热休克蛋白70对人肺腺癌A549细胞上皮间质转化的影响 被引量:2

摘要 目的:探讨热休克蛋白70(HSP70)对A549细胞上皮间质转化(EMT)进程的影响。方法:筛选替普瑞酮诱导HSP70表达的最佳作用条件。利用TGF-β刺激A549细胞建立EMT模型。将细胞分为对照组、TGF-β组、替普瑞酮组、TGF-β+替普瑞酮组,TGF-β的作用浓度为5μg/L。应用Western blot、划痕实验检测各组A549细胞中EMT相关蛋白E-cadherin、Vimentin的表达和细胞迁移能力。结果:选择50μmol/L的替普瑞酮用于实验;TGF-β促使A549细胞E-cadherin蛋白表达降低,Vimentin蛋白表达升高,迁移能力增强;替普瑞酮促使A549细胞E-cadherin蛋白表达降低,Vimentin蛋白表达降低,迁移能力下降;替普瑞酮可拮抗TGF-β促A549细胞EMT的作用(P<0.05)。结论:HSP70高表达可抑制A549细胞EMT进程和细胞的迁移。 Aim:To investigate the effects of heat shock protein 70(HSP70)on the process of epithelial-mesenchymal transition(EMT)in A549 cells.Methods:The optimal conditions for teprenone was screened to induce the expression of HSP70.TGF-βwas used to stimulate A549 cells to establish the EMT model.The cells were allocated into control group,TGF-βgroup,teprenone group,TGF-β+teprenone group and the concentration of TGF-βwas 5μg/L.Western blot and Scratch test were respectively used to detect the expression of EMT-related proteins E-cadherin and Vimentin as well as cell migration ability in each group.Methods:Teprenone at 50μmol/L was selected for the experiment.TGF-βreduced the expression of E-cadherin,increased the expression of Vimentin in A549 cells,and enhanced the migration ability.The expression of E-cadherin and Vimentin in A549 cells were decreased by teprenone,and the migration ability was decreased.Teprenone could antagonize the EMT effect of TGF-βin A549 cells(P<0.05).Conclusion:The high expression of HSP70 can inhibit the EMT process and cell migration ability of A549 cells.
