摘要 目的:建立UPLC-DAD法测定款冬花中克氏千里光碱的含量。方法:采用沃特世CORTECS UPLC C_(18)(2.1 mm×100 mm,1.6μm)色谱柱,流动相为乙腈-0.1%磷酸水溶液(15∶85);检测波长220 nm;柱温:30℃;进样量:1μL。结果:克氏千里光碱在2.048~81.82μg·mL^(-1)浓度范围内与峰面积呈现良好的线性关系(r=1.0000);平均加样回收率分别为89.46%,RSD分别为2.80%;10批样品中克氏千里光碱的含量为0.00236~0.00534%。结论:该方法简便、快速,可用于款冬花中克氏千里光碱的含量测定。 Objective:To establish an UPLC-DAD method for quantitative analysis of senkirkine in Farfarae Flos.Methods:The assay was performed on an Waters CORTECS UPLC C_(18)(2.1 mm×100 mm,1.6μm)column and the sample was eluted with a mobile phase consisted of acetonitrile-0.1%phosphoric acid aqueous solution(15∶85).The detection wavelength was set at 220 nm,and the column temperature was 30℃.Injection volume:1μL.Results:The linear range of senkirkine was 2.048-81.82μg·mL^(-1)(r>1.0000).The average recovery was 89.46%with RSD of 2.80%.The contents of senkirkine in Farfarae Flos were 0.00236-0.00534%.Conclusion:The proposed method is simple and rapid,can be used as content determination of senkirkine in Farfarae Flos.
