邻苯二甲酸二(2-乙基己基)酯亚慢性染毒对wistar大鼠血液学及血生化指标的影响 被引量:1

Subchronic exposure of bis(2-ethylhexyl)phthalate to blood cells and blood cells of wistar rats and the impact of biochemical indicators

摘要 目的观察邻苯二甲酸二(2-乙基己基)酯(DEHP)染毒90 d对大鼠血液学及血生化指标的影响。方法将健康SPF级Wistar大鼠80只,雌雄各半,按体重随机分为对照组和3个DEHP染毒组(500、100、20 mg/kg)各20只,经口90 d染毒后,采集大鼠下腔静脉血,进行血常规、血生化常规检测。结果血液学检测结果显示,500mg/kg DEHP染毒组大鼠红细胞数、白细胞数、血红蛋白数量下降,与对照组比较差异有统计学意义(F_(雄)=1.891、1.641、2.328,F雌=2.011、2.392、3.028;均P<0.05)。大鼠白细胞分类中,500mg/kg DEHP染毒组雄、雌性大鼠单核细胞计数均升高,与对照组比较差异均有统计学意义(F_(雄)=2.418,F雌=1.436;均P<0.05);DEHP各剂量组淋巴细胞、中性粒细胞、嗜酸性细胞、嗜碱性细胞数量与对照组比较,差异均无统计学意义(均P>0.05)。生化指标检测结果显示,雄性大鼠中500 mg/kg DEHP剂量组血生化AST、TP、BUN水平升高,与对照组比较差异有统计学意义(F_(雄)=5.825、2.935、3.525,均P<0.05);500、100 mg/kg DEHP染毒组血生化ALT、Crea水平升高,与对照组比较差异有统计学意义(F_(雄)=9.769、3.633,均P<0.05);500 mg/kg DEHP染毒组血生化ALT、AST、BUN升高,与20 mg/kg DEHP染毒组比较差异均有统计学意义(F=1.186、0.678、0.036,P<0.05)。雌性大鼠中500 mg/kg DEHP剂量组血生化AST、TP、BUN、Crea水平升高,与对照组比较差异均有统计学意义(F雌=4.656、2.848、2.405、2.614,均P<0.05);500、100 mg/kg DEHP染毒组血生化ALT水平升高,与对照组比较差异有统计学意义(F雌=11.776,P<0.05)。结论500~20mg/kg剂量下亚慢性DEHP暴露对wistar大鼠血液学和血生化具有潜在危害。 Objective To observe the effects of bis(2-ethylhexyl)phthalate(DEHP)exposure for 90 days on hematology and serum biochemistry in rats.Methods Eighty healthy SPF Wistar rats,half male and half female,were randomly divided into control group and 3 DEHP-exposed groups(500,100,20 mg/kg)according to their body weight.After oral administration for 90 days,they were collected.The blood of the inferior vena cava of rats was tested for routine blood and biochemical routine.Results The results of hematology test showed that the number of red blood cells,white blood cells,and hemoglobin in the 500 mg/kg DEHP exposure group decreased,and the difference was statistically significant compared with the control group(Fmale=1.891,1.641,2.328,Ffemale=2.011,2.392,3.028,all P<0.05).In the classification of white blood cells in rats,the mononuclear cell counts of male and female rats in the 500 mg/kg DEHP exposure group increased,and the difference was statistically significant compared with the control group(Fmale=2.418,Ffemale=1.436,all P<0.05);Compared with the control group,the number of lymphocytes,neutrophils,eosinophils,and basophils in each dose group of DEHP was not statistically significant(P>0.05).The biochemical index test results showed that the serum biochemical AST,TP,and BUN levels in the 500 mg/kg DEHP dose group increased in male rats,which were significantly different from those in the control group(Fmale=5.825,2.935,3.525,all P<0.05);The serum biochemical ALT and Crea levels in the 500,100 mg/kg DEHP exposure group increased,and the difference was statistically significant compared with the control group(Fmale=9.769,3.633,all P<0.05);500 mg/kg DEHP exposure Serum biochemical ALT,AST and BUN increased in the poison group,which were significantly different from those in the 20 mg/kg DEHP group(F=1.186,0.678,0.036,all P<0.05).The levels of serum biochemical AST,TP,BUN,and Crea in the 500 mg/kg DEHP dose group increased in female rats,and the differences were statistically significant compared with those in the control group(Ffemale=4.656,2.848,2.405,2.614,all P<0.05);The level of serum biochemical ALT in the 500,100 mg/kg DEHP exposure group increased,and the difference was statistically significant compared with the control group(Ffemale=11.776,P<0.05).Conclusion Long-term exposure to DEHP at a dose of 500-20 mg/kg is potentially harmful to hematology and serum biochemistry in wistar rats.
