镁对下肢动脉硬化闭塞症大鼠股-腘动脉内皮lncRNA-sONE的影响 被引量:1

Effect of Magnesium on LncRNA-Sone of Femoropopliteal Artery Endothelial in Rats with Lower Extremity Arteriosclerosis Obliterans

摘要 目的研究镁对下肢动脉粥样硬化闭塞性大鼠股-月国动脉内皮细胞lncRNA-sONE的影响。方法 100只动脉粥样硬化闭塞性大鼠随机分为模型组22例和实验组22例。随机选取100只SD大鼠,纵向切开腹股沟,暴露股-月国动脉,不进行剪切和剥离。其他为假手术组25例。实验组大鼠灌胃30 mg/(kg·d)^(-1)镁溶液,假手术组和模型组大鼠灌胃等量生理盐水。各组干预时间均为2周。自动生化分析仪检测各组大鼠血脂水平,自动生化分析仪检测各组大鼠血清总胆固醇(TC)和甘油三酯(TG)水平。结果镁可影响脂质过氧化,保护细胞膜,维持线粒体功能,拮抗Ca^(2+)内流,影响lncRNA-sONE的表达。实验组与模型组比较差异有统计学意义(P<0.05)。结论镁可有效调节动脉粥样硬化闭塞性大鼠高脂血症水平,改善大鼠股月国动脉内皮中lncRNA-sONE的功能,修复内皮损伤。其作用机制可能与抑制股-月国动脉lncRNA-sONE的表达有关。 Objective To study the effect of magnesium on the lncRNA-sONE of femoropopliteal artery endothelial in rats with lower extremity arteriosclerosis obliterans.Methods A total of 100 rats with atherosclerosis obliterans were randomly divided into model group(n=22)and experimental group(n=22).Another 100 SD rats were randomly selected to have the groin cut longitudinally to expose the femoro-popliteal artery without clipping or dissection.The other operations were sham operation group(n=25).The rats in the experimental group were intragastrically given 30 mg/(kg·d)^(-1) magnesium solution,while the rats in the sham operation group and the model group were intragastrically given the same amount of normal saline.The intervention in all groups lasted for 2 weeks.Blood lipid levels of rats in each group were detected by automatic biochemical analyzer,and serum total cholesterol(TC)and triglyceride(TG)levels of rats in each group were detected by automatic biochemical analyzer.Results Magnesium can affect lipid peroxidation,protect cell membrane,maintain mitochondrial function,antagonize Ca2+influx,and affect the expression of lncRNA-sONE.The differences between the experimental group and the model group were statistically significant(P<0.05).Conclusions Magnesium can effectively regulate the level of hyperlipidemia in rats with arteriosclerosis obliterans,improve the function of lncRNA-sONE in the femoropopliteal artery endothelial of the rats,and repair the endothelial injury.The mechanism of action may be related to its inhibition of the expression of lncRNA-sONE in the femoropopliteal artery.
