不同培养温度及孢子稀释液对黑牛肝菌孢子萌发的影响 被引量:2

The Effects of Different Culture Temperature and Dilution on Spore Germination of Phlebopus portentosus

摘要 通过调整培养温度、浸泡时间,以及孢子稀释液的pH、添加碳源、氮源等条件,研究各因素对黑牛肝菌(Phlebopus portentosus)担孢子萌发的影响。结果表明,25℃、30℃、35℃下培养时平均萌发菌落数显著高于15℃、20℃下培养,并以30℃为最优;涂布前将孢子以无菌水浸泡24 h,能显著提高萌发菌落数;调节孢子稀释液pH为6,在稀释液中添加1%葡萄糖、添加0.1%酒石酸铵或尿素,均能在一定程度上提高萌发菌落数。 Different culture temperature, soaking time, and the pH, carbon source, nitrogen source of dilution was tested to study their effects on the gemination of Phlebopus portentosus spore. The results showed that, under the culture temperature of 25℃,30℃ and 35℃, the germination numbers are significantly higher than those cultured under 15℃ and 20℃, and 30℃ is the optimal culture temperature. After soaking in sterile water for 24 h, the germination number can be significantly raised. Adjusting the pH of spore diluent to 6, adding 1% glucose and 0.1% ammonium tartrate or urea in the diluent could increase the number of germinating colony to some extent.
