摘要
【目的】为了探讨CYP19基因在牦牛睾丸及附睾不同部位的表达差异性.【方法】应用免疫组织化学、qRT-PCR方法、Western-blot等检测方法,研究成年牦牛睾丸及附睾头、体、尾等4个组织中CYP19基因的表达和定位.【结果】免疫组化结果显示在睾丸支持细胞、间质细胞和生殖细胞中P450arom蛋白均有较强表达,在附睾头、体、尾的上皮细胞和平滑肌细胞均有较弱表达.蛋白印迹结果显示附睾头表达最高,附睾尾、附睾体、睾丸表达依次递减(P<0.05).mRNA水平结果表明睾丸中表达量最高,附睾尾、附睾头、附睾体表达依次递减(P<0.01).【结论】以上结果提示CYP19基因在睾丸及附睾头、体、尾中表达有差异,这一结果为进一步研究CYP19基因对牦牛睾丸及附睾发育作用奠定了基础.
【Objective】To explore the difference in CYP19 expression among different parts of testis and epididymis of Bos grunniens.【Method】The expression and localization of CYP19 in the testis,caput,corpus,and cauda of adult Bos grunniens were studied by the ways of immunohistochemistry,qRT-PCR and Western blot method.【Result】The results by immunohistochemistry showed that P450arom protein was strongly expressed in Sertoli cells,Leydig cells,and germ cells of the testis,and weakly expressed in epithelial cells and smooth muscle cells of the caput,corpus and cauda.Western-blot results showed that the expression in caput was the highest,and decreased successively in cauda,corpus,and testis(P<0.05).In terms of the mRNA level,it was showed that the expression was the highest in the testis,and decreased successively in the cauda,caput,and corpus(P<0.01).【Conclusion】Taken together,it was indicated that the expression of CYP19 gene was difference in the testis,caput,corpus and cauda,and the findings laid a foundation for further study on the role of CYP19 gene in the development of testis and epididymides of B.grunniens.
