低氧环境下甘肃鼢鼠延髓中c-Fos和HIF-1α的表达模式分析 被引量:1

摘要 【目的】低氧环境可造成中枢神经系统的损伤,延髓可通过对自身的调控来应对低氧协迫.本研究利用甘肃鼢鼠自身的低氧适应能力来分析其与SD大鼠延髓中不同核团的低氧适应机制.【方法】采用免疫组织化学法,在常氧、急性低氧和间歇性慢性低氧条件下,分析延髓孤束核(NTS)、灰质(GC)及蓝斑核(LC)中c-Fos和HIF-1α的表达.【结果】在常氧条件下甘肃鼢鼠NTS、LC内c-Fos表达极显著高于SD大鼠;间歇性慢性低氧条件下甘肃鼢鼠三核团c-Fos表达均极显著高于SD大鼠;其他条件下差异不明显.3种条件下,甘肃鼢鼠三核团HIF-1α表达量均极显著高于SD大鼠.间歇性慢性低氧条件下,雄性甘肃鼢鼠3种核团及急性低氧条件下孤束核(NTS)HIF-1α表达显著高于雌性,c-Fos表达极显著高于雌性.【结论】甘肃鼢鼠延髓系统较SD大鼠有更强的低氧适应能力,且雌性甘肃鼢鼠较雄性低氧耐受性强. 【Objective】Hypoxic environment can cause damage to the central nervous system,and the medulla oblongata can respond to hypoxic stress by regulating itself.In this paper,we used Myspalax cansus′s own ability to adapt to hypoxia to analyze the mechanism of adaptation to hypoxia in different nuclei in the medulla oblongata of SD rats.【Method】Using immunohistochemistry,the nucleus tractus solitarius(NTS),gray matter(GC)and nucleus coeruleus(LC)were analyzed for c-Fos and expression of HIF-1αunder normoxia,acute hypoxia and intermittent chronic hypoxia.【Result】Under normoxic conditions,the expression of c-Fos in M.cansus NTS and LC was significantly higher than that in SD rats.Under intermittent chronic hypoxia conditions,the expression of c-Fos in the trinucleus of M.cansu was significantly higher compared with SD rats.The difference was not obviously under other conditions.Under the three conditions,the expression of HIF-1αin the trinucleus of M.cansu was significantly higher than that of SD rats.Under intermittent chronic hypoxia conditions,the three nuclei of male M.cansu and the nucleus NTS HIF-1αexpression were significantly higher than females under acute hypoxia conditions,and the expression of c-Fos was extremely significantly higher than females.【Conclusion】The medulla oblongata system of M.cansu has a stronger ability to adapt to hypoxia than SD rats,and the female is more tolerant to hypoxia than males.
