摘要
目的建立桂皮醛和柴胡皂苷a的HPLC同时测定方法,评价桂枝柴胡颗粒的质量。方法含量测定使用Agilent SB C 18色谱柱(250 mm×4.6 mm,5μm),流动相为乙腈(A)-0.1%磷酸溶液(B,v/v),梯度洗脱程序为:0~10 min,5%~10%A;10~30 min,10%~40%A;30~35 min,40%~75%A;35~40 min,75%~5%A;流速1.0 mL·min^(-1),检测波长为210 nm(柴胡皂苷A),290 nm(桂皮醛),柱温30℃,进样量10μL;共测定10批桂枝柴胡颗粒。结果10批样品中,柴胡皂苷a的含量为0.4558~0.6936 mg·g^(-1),桂皮醛的含量为0.2574~0.4097 mg·g^(-1)。结论本研究建立了桂枝柴胡颗粒两种指标性成分含量同时测定方法,该方法快速、准确,能够较好地对桂枝柴胡颗粒进行质量评价。
OBJECTIVE To evaluate the quality of GuiZhi ChaiHu Granules,by means of establishing simultaneous determination method for saikoside a and cinnamaldehyde.METHODS Content determination used Agilent SB C 18 column(250 mm×4.6 mm,5μm),the mobile phase comprised of acetonitrile(A)-0.1%phosphoric acid solution(v/v,B),the program of gradient elution was 0-10 min,5%-10%A;10-30 min,10%-40%A;30-35 min,40%-75%A;35-40 min,75%-5%A;the flow rate was 1.0 mL·min^(-1),the detection wavelength was 210 nm(for saikoside a)and 290 nm(for cinnamaldehyde),the column temperature was 30℃,the injection volume was 10μL;10 batches of samples were analyzed.RESULTS In 10 batches of samples,content of saikoside a was 0.4558-0.6936 mg·g^(-1),that of cinnamaldehyde was 0.2574-0.4097 mg·g^(-1).CONCLUSION A simultaneous determination method for two components in GuiZhi ChaiHu Granules was established in this study,which was fast and accurate for better evaluation of the granule′s quality.
