基于网络药理学和代谢组学探讨滋阴安神方治疗失眠作用机制 被引量:6

Mechanism of Prescriptions for Nourishing Yin and Tranquilizing Mind in Treating Insomnia Based on Network Pharmacology and Metabonomics

摘要 目的:基于网络药理学以及代谢组学,以酸枣仁汤与百合地黄汤为例,探讨滋阴安神方治疗失眠的分子机制。方法:借助网络药理学,以相似性显著的成分为基础,预测两方作用靶点,结合液质联用技术分析药物干预后大鼠血清,综合分析两方核心靶点集合、核心作用通路集合。结果:两方共同作用靶点11个、作用通路11条,主要通过地黄苷a、梓醇、酸枣仁皂苷a1等29个小分子发挥治疗失眠作用。结论:两方作用核心靶点在肝脏中表达最多,参与神经活性配体-受体相互作用、外源物质代谢、胆汁分泌、突触囊泡循环等多个信号通路。揭示了滋阴安神类复方治疗失眠的可能疗效机制,为下一步从“肝”入手治疗失眠提供了新的研究思路。 Objective:To explore the molecular mechanism of prescriptions for nourishing yin and tranquilizing mind in treating insomnia based on network pharmacology and metabonomics,taking Suanzaoren Decoction(酸枣仁汤)and Baihe Dihuang Decoction(百合地黄汤)as examples.Methods:With the help of network pharmacology,the targets of the two prescriptions were predicted based on the components with significant similarity.The serum of rats after drug intervention was analyzed by liquid/mass spectrometry technology,and the core targets and core action pathways were comprehensively analyzed.Results:There were total 11 common targets and 11 action pathways in the two prescriptions,mainly through 29 small molecules of rehmaionoside a,catalpol,jujuboside a1 and so on to play a therapeutic effect on insomnia.Conclusions:The core targets which are most expressed in the liver and are involved in neuroactive ligand-receptor interactions,exogenous substance metabolism,bile secretion,synaptic vesicle cycle,etc.It reveals the efficacy mechanism of prescriptions for nourishing yin and tranquilizing mind in treating insomnia and provides new research ideas for the further treatment of insomnia from“liver”.
