Screening of Extraction Methods and Content Determination of Total Flavonoids in Wild Macleaya Cordata

摘要 为了探究不同提取方法对野生博落回总黄酮含量的影响和分析不同地区不同生长部位的野生博落回的总黄酮含量差异,采集福建省南平市光泽县和福建省三明市明溪县两地的野生博落回样品,采用甲醇提取法、超声甲醇提取法、超声甲醇补足提取法、超声乙醇提取法和乙醇提取法等5种不同的提取方法对博落回中总黄酮进行提取,利用NaNO_(2)·Al(NO_(3))_(3)比色法对总黄酮的含量进行测定,并利用SPSS 18.0对数据进行统计与分析。结果表明,超声甲醇补足提取法的提取效果最好,博落回叶的总黄酮含量和提取率最高。光泽和明溪两地的野生博落回叶中的总黄酮含量和总黄酮提取率均高于根和茎,光泽野生博落回样品各部位中总黄酮含量和总黄酮提取率比明溪的略高,但无显著差异。 In order to explore the effects of different extraction methods on the total flavonoid content of wild Macleaya cordata and to analyze the difference of total flavonoid content in different growth parts of wild Macleaya cordata in different regions,the samples of wild Macleaya cordata from Guangze County of Nanping City and Mingxi County of Sanming City in Fujian Province were collected,and five different extraction methods,including the methanol extraction,ultrasonic-methanol extraction,ultrasonic-methanol supplement extraction,ultrasonic-ethanol extraction,and ethanol extraction,were used to extract the total flavonoids from Macleaya cordata.Then,NaNO_(2)·Al(NO_(3))_(3) colorimetry was used to determine the content of total flavonoids,and SPSS18.0 was used for the data statistics and analysis.The results showed that the ultrasonic-methanol supplemental extraction method had the best extraction effect,and the content and extraction rate of total flavonoids were the highest.The content and extraction rate of total flavonoids in the leaves of wild Macleaya cordata in Guangze and Mingxi were higher than those in the roots and stems.The content and extraction rate of total flavonoids in different parts of the samples of wild Macleaya cordata in Guangze were slightly higher than that in Mingxi,but there was no significant difference.
