乳腺癌中ApoM差异表达及其对HER2阳性且HR阳性乳腺癌细胞株的生物学影响 被引量:2

Differential expression of ApoM in breast cancer and its biological effects on HER2-positive and HR-positive breast cancer cell lines

摘要 目的探讨载脂蛋白M(ApoM)在乳腺癌中差异表达及其对人表皮生长因子受体2(HER2)阳性且激素受体(HR)阳性的乳腺癌细胞株BT-474和ZR-75-1的生物学影响。方法检测55例乳腺癌患者癌组织和癌旁组织中ApoM表达水平。应用慢病毒载体构建ApoM过表达体系并转染HER2阳性且HR阳性的乳腺癌细胞株BT-474和ZR-75-1,并分为ApoM过表达(ApoM-OE)组和空病毒对照组。采用CCK-8细胞增殖实验、细胞划痕实验和Transwell小室实验分析ApoM对HER2阳性且HR阳性BT-474和ZR-75-1细胞增殖、迁移和侵袭的影响。结果癌组织中ApoM的mRNA水平较癌旁组织降低(P<0.05)。BT-474和ZR-75-1细胞中ApoM mRNA表达水平低(P<0.05或P<0.01)。转染后,BT-474和ZR-75-1细胞ApoM-OE组ApoM mRNA表达水平升高(P<0.01)。ZR-75-1细胞中ApoM-OE组在24、48、72 h细胞增殖率下降(P<0.05或P<0.01)。BT-474细胞中ApoM-OE组在24、48 h的划痕面积百分比升高(P<0.05或P<0.01)。BT-474和ZR-75-1细胞中ApoM-OE组侵袭细胞数下降(P<0.05)。结论 ApoM在乳腺癌组织中表达降低,过表达ApoM可以抑制HER2阳性且HR阳性乳腺癌细胞增殖、迁移及侵袭。 Objective To investigate differential expression of apolipoprotein M(ApoM)in breast cancer and its biological effects on human epidermal growth factor receptor 2(HER2)positive and hormone receptor(HR)positive breast cancer cell lines BT-474 and ZR-75-1.Methods The expression levels of ApoM in cancer tissues and tumor-adjacent tissues of 55 patients with breast cancer were detected.Lentiviral vector was used to construct ApoM overexpression system and transfected HER2-positive and HR-positive breast cancer cell lines BT-474 and ZR-75-1,which were divided into ApoM overexpression group(ApoM-OE group)and empty virus control group.CCK-8 cell proliferation assay,cell scratch assay and Transwell chamber assay were used to analyze the effects of ApoM on the proliferation,migration and invasion of HER2-positive and HR-positive BT-474 and ZR-75-1 cells.Results The mRNA level of ApoM in the cancer tissues was lower than that in the tumor-adjacent tissues(P<0.05).The mRNA expression levels of ApoM in BT-474 and ZR-75-1 cells were lower(P<0.05 or P<0.01).After transfection,ApoM mRNA expression levels of BT-474 and ZR-75-1 cells were increased in ApoM-OE group(P<0.01).In ZR-75-1 cells,the cell proliferation rate of ApoM-OE group was decreased at the 24th,48th and 72nd hour(P<0.05 or P<0.01).The percentage of scratch area increased at the 24th and 48th hour in BT-474 cells(P<0.05 or P<0.01).The cell number of ApoM-OE group in BT-474 and ZR-75-1 cells was decreased(P<0.05).Conclusion The expression of ApoM is decreased in breast cancer tissues,and overexpression of ApoM can inhibit the proliferation,migration and invasion of HER2-positive and HR-positive breast cancer cells.
