摘要
藏香猪是一种生长在高原上的具有独特特性的本土猪种,具有抗氧化应激能力强的特点。本研究使用超声波辅助复合酶酶解的方法处理藏香猪血液总蛋白,利用超滤离心得到具有高活性的酶解产物组分。研究表明,使用复合酶(胰蛋白酶∶木瓜蛋白酶∶胰凝乳蛋白酶,1∶1∶1)按照50 U/100 mL的添加量处理猪血总蛋白6 h可使酶解产物的水解度达到最大,约为23.6%,此时酶解产物的DHHP清除能力最高,约为63.4%。短时低功率的超声波辅助处理可使酶解产物的DHHP清除能力提升至71.3%。80%以上的具有DHHP清除能力的酶解产物集中在小于1 KDa的小肽中。本研究使用的超声波辅助复合酶酶解法适用于藏香猪血液中抗氧化肽的提取。对比藏香猪和长白猪血液酶解产物的水解度和抗氧化活性,发现二者水解度相当,但是藏香猪血液的酶解产物对DHHP的清除能力比长白猪高20%左右。
Tibetan pig is a native pig breed with unique characteristics growing on plateau,which has strong ability of antioxidative stress.In this study,ultrasound assisted enzymatic hydrolysis was used to treat the total protein of Tibetan pig blood,and the enzymatic hydrolysate with high activity was obtained by ultrafiltration centrifugation.The results showed that the hydrolysis degree of the hydrolysate was the highest,about 23.6%,and the DHHP scavenging ability of the hydrolysate was the highest,about 63.4%,when the total protein of pig blood was treated with 50 U/100 mL compound enzyme(trypsin:papain:chymotrypsin,1∶1∶1)for 6 h.The DHHP scavenging ability of enzymatic hydrolysates was enhanced to 71.3%by short-time and low-power ultrasonic treatment.More than 80%of the hydrolysates with DHHP scavenging ability were concentrated in small peptides,which were lighter than 1 kDa.These results revealed that the ultrasonic assisted enzymatic hydrolysis was suitable for the extraction of antioxidant peptides from Tibetan pig blood.Comparing the degree of hydrolysis and antioxidant activity of the hydrolysates from Tibetan and Landrace pig blood,it was found that the degree of hydrolysis of the two hydrolysates was the same,but the DHHP clearing ability of the hydrolysates from Tibetan pigs Blood was about 20%higher than that of landrace pig blood.
