银香解毒膏抑制金黄色葡萄球菌诱发皮肤炎性反应的实验研究

Experimental study of Yinxiang Jiedu ointment in inhibiting skin inflammation induced by Staphylococcus aureus

目的:探讨银香解毒膏降低金黄色葡萄球菌对皮肤的损伤程度以及抑制其炎性因子的表达水平,初步阐述银香解毒膏保护金黄色葡萄球菌感染后皮肤损伤的相关的炎性分子表征。方法:使用“胶带剥脱法”建立小鼠金黄色葡萄球菌感染模型,小鼠感染24 h后使用银香解毒膏莫匹罗星膏、赋形剂膏进行涂抹感染部位,实验中统一用量。按照实验设计时间分别在涂抹药物治疗后的第3天和第5天时皮肤取样,一部分进行OCT冰冻包埋制作HE染色,一部分进行总RNA提取对相关炎性因子进行PCR。结果:银香解毒膏可以很好地保护被金黄色葡萄球菌感染的皮肤损伤,防止皮肤损伤扩大化,并促进皮肤损伤部位快速愈合,减低皮肤损伤部位IL-6、TNF-α、IL-1β、IL-4炎性因子水平。结论:本研究表明银香解毒膏可以降低金黄色葡萄球菌对皮肤的损伤程度以及抑制其炎性因子的表达水平,揭示了银香解毒膏对金黄色葡萄球菌诱导的皮肤损伤有显著的保护作用。

Objective To investigate whether Yinxiang Jiedu ointment can reduce the damage of Staphylococcus aureus to the skin and inhibit the expression of inflammatory factors in the skin,and to preliminarily explain that Yinxiang Jiedu ointment protects the skin after Staphylococcus aureus infection Molecular characterization of related inflammation.Method Use the“tape stripping method”to establish a mouse model of Staphylococcus aureus infection.24 hours after the mice were infected,use Yinxiang Jiedu ointment,mupirocin ointment,and excipient ointment to smear the infected area,and uniform dosage in the experiment.According to the experimental design time,the skin was sampled on the 3rd and 5th days after the drug treatment,some of them were frozen and embedded in OCT,HE staining was made,some were extracted for total RNA,and the relevant inflammatory factors were subjected to PCR.Results We found that Yinxiang Jiedu ointment can protect the skin lesions infected by Staphylococcus aureus very well,prevent the skin lesions from expanding,and promote the rapid healing of skin lesions,and reduce the IL-6,TNF-α,IL-1β,IL-4 inflammatory factor level.Conclusion Our research shows that Yinxiang Jiedu ointment can reduce the damage of Staphylococcus aureus to the skin and inhibit the expression of inflammatory factors.In short,our findings reveal that Yinxiang Jiedu ointment can induce Staphylococcus aureus the skin damage has a significant protective effect.
