基于GC-MS的九香虫脂肪油入血成分分析 被引量:3

摘要 目的:研究九香虫脂肪油给大鼠灌胃后的入血成分,初步揭示九香虫脂肪油体内潜在药效物质基础。方法:取大鼠12只雌雄各半,随机分为空白组及给药组,给药组以九香虫脂肪油灌胃,空白组灌胃等量的生理盐水。采集血液后,将处理好的血浆采用GC-MS(气相色谱-质谱联用仪)测定大鼠血浆中九香虫脂肪油入血的成分。结果:对比空白血浆、含药血浆和九香虫脂肪油3种离子流图,指认了5个入血原型成分[3,5-二(1,1-二甲基乙基)苯酚、软脂酸甲酯、油酸甲酯、14-甲基十五烷酸甲酯、乙酸(6Z,13Z)-十八碳烯酯]和3个可能为九香虫脂肪油体内代谢物,即烯丙基草酸壬酯、(11,14)-二十碳二烯酸甲酯、四环素[7.1.0(1,6)·0(4,9)·0(8,10)]癸烷。结论:本实验初步确定了九香虫脂肪油的血中移行成分,为阐明其发挥药效的潜在物质基础提供了实验依据。 Objective:To reveal the underlying substance base of aspongopus′s efficacy through studying the blood components after rats were given gastric administration aspongopus fatty oil.Methods:6 male and 6 female rats were randomly divided in blank group and administration group,and rats in administration group were intragastrically administered aspongopus fat oil while rats in blank group were given equal volume of saline.Medicinal composition in plasma was identified by GC-MS(gas chromatography-mass spectrometry).Results:It was found that 3,5-bis(1,1-dimethylethyl)phenol,palmitic methy1 ester,methy1 oleate,14-methyl pentadecanoate methy1 ester,acetic acid(6Z,13Z)-octadecene ester were metabolites of aspongopus fatty oil,and allyl oxalic acid pelargonic ester,11,14-eicosadienoic acid methy1 ester,tetracycline[7.1.0(1,6)·0(4,9)·0(8,10)]decane may be the metabolites of the aspongopus fatty oils through comparing ion flow diagrams of blank plasma,drug containing plasma and aspongopus fatty oil.Conclusion:The constituents absorbed into blood after intragastric administration of aspongopus fatty oil are identified in this study,and provides experimental basis for elucidating the potential material basis of efficacy of aspongopus fatty oil.
