TAB3、CEBPD在卵巢癌组织和卵巢癌细胞中的表达及其相关性

Expression and Correlation of TAB3 and CEBPD in Ovarian Cancer Tissue and Ovarian Cancer Cells

目的探讨TAB3、CEBPD在卵巢癌组织和卵巢癌细胞中的表达及其相关性。方法收集51例临床卵巢癌病理标本和对应的癌旁组织标本,采用qRT-PCR及western blot方法检测卵巢癌组织及对应的癌旁组织中TAB3、CEBPD的表达情况,同时采用qRT-PCR及western blot方法检测正常卵巢细胞和卵巢癌细胞株中TAB3、CEBPD的表达,分析TAB3、CEBPD的相关性。结果荧光定量PCR结果发现,卵巢癌组织中TAB3 mRNA的表达明显高于癌旁组织(P<0.001),而CEBPD mRNA的表达明显低于癌旁组织(P<0.05);Western blot结果发现TAB3蛋白在62.74%(32/51)的卵巢癌组织中呈过表达,CEBPD蛋白在60.78%(31/51)的卵巢癌组织中呈低表达(P<0.05)。卵巢癌细胞株中的TAB3 mRNA及蛋白表达均明显高于正常卵巢细胞株,CEBPD mRNA及蛋白表达明显低于正常卵巢细胞株。TAB3、CEBPD在卵巢癌组织中的表达呈负相关(γ=-0.2853,P<0.05;γ=-0.3091,P<0.05)。结论在卵巢癌组织及细胞中TAB3基因呈高表达,而CEBPD基因呈低表达。在卵巢癌组织中TAB3、CEBPD基因的表达存在负相关性。

Objective To investigate the expression and correlation of TAB3 and CEBPD in ovarian cancer tissues and cells.Methods The expressions of TAB3 and CEBPD were detected by QRT PCR and Western blot in 51 cases of ovarian cancer and corresponding adjacent tissues.The expressions of TAB3 and CEBPD in normal ovarian cells and ovarian cancer cell lines were detected by QRT PCR and Western blot.Results The expression of TAB3 mRNA in ovarian cancer tissues was significantly higher than that in adjacent tissues(P<0.001),while the expression of CEBPD mRNA in ovarian cancer tissues was significantly lower than that in adjacent tissues(P<0.05).Western blot showed that TAB3 protein was overexpressed in 62.74%(32/51)of ovarian cancer tissues,and CEBPD protein was 60.78%(31/51)There was a low expression in ovarian cancer(P<0.05).The expression of TAB3 mRNA and protein in ovarian cancer cell line was significantly higher than that in normal ovarian cell line,and the expression of CEBPD mRNA and protein was significantly lower than that in normal ovarian cell line.The expression of TAB3 and CEBPD was negatively correlated in ovarian cancer tissue(γ=-0.2853,P<0.05,γ=-0.3091,P<0.05).Conclusion TAB3 gene is highly expressed in ovarian cancer tissues,while CEBPD gene is low expression.There is a negative correlation between the expression of TAB3 and CEBPD genes in ovarian cancer tissues and cells.
