鼠双微体基因2以及核受体结合SET结构域蛋白2在乳腺癌组织的表达及其临床意义 被引量:2

Expression and significance of murine double minute 2 and nuclear receptor binding SET domain-protein 2 in breast carcinoma tissues

摘要 目的:探讨鼠双微体基因2(MDM2)和核受体结合SET结构域蛋白2(NSD2)在乳腺癌组织中的表达及意义。方法:收集2014年7月至2020年1月广东医科大学附属医院103例乳腺癌组织和配对癌旁组织,采用免疫组织化学方法检测组织中MDM2蛋白以及NSD2蛋白的表达,结合临床资料行χ^(2)检验相关性分析。结果:MDM2蛋白在乳腺癌组织中表达率为69.90%(72/103),显著高于配对癌旁组织中表达率10.68%(11/103),两者差异有统计学意义(χ^(2)=75.083,P<0.01),MDM2表达水平与乳腺癌的TNM分期、组织学分级、肿瘤大小、淋巴结转移明显相关(χ^(2)=5.501、8.724、4.127、5.415,P<0.05)。NSD2蛋白在乳腺癌组织中表达率为71.85%(74/103),显著高于配对癌旁组织中表达率11.65%(12/103),两者差异有统计学意义(χ^(2)=76.731,P<0.01),NSD2蛋白表达水平与乳腺癌的淋巴结转移、组织学分化程度、TNM分期明显相关(χ^(2)=7.206、5.793、4.995,P<0.05),差异具有统计学意义。结论:MDM2蛋白以及NSD2蛋白异常高表达可能参与乳腺癌发生发展与转移,检测MDM2以及NSD2有助于判价乳腺癌的恶性程度、判断乳腺癌转移潜能。 Objective To explore the expression and significance of murine double minute 2(MDM2)and nuclear receptor binding SET domain-protein 2(NSD2)in breast carcinoma tissues.Methods The expression of MDM2 and NSD2 proteins in 103 cases of breast cancer tissues and matched non-cancerous adjacent breast tissues was collected from Affiliated Hospital of Guangdong Medical University from July 2014 to January 2020.The expression of MDM2 and NSD2 proteins was detected by envision immunohistochemical method,and the correlation analysis was performed by chi-square test combined with clinical data.Results The expression rate of MDM2 protein in breast cancer tissues was 69.90%(72/103),which was significantly higher than that in matched non-cancerous adjacent breast tissues[10.68%(11/103)]with the difference being statistically significant(χ^(2)=75.083,P<0.01).The expression of MDM2 was significantly correlated with tumor node metastasis stage(TNM stage),histological stage,tumor size and lymph node metastasis of breast cancer(respectively,χ^(2)=5.501,8.724,4.127,5.415,P<0.05).The expression rate of NSD2 protein in breast cancer tissue was 71.85%(74/103),which was significantly higher than that in matched non-cancerous adjacent breast tissue[11.65%(12/103)]with the difference being statistically significant(χ^(2)=76.731,P<0.01).The expression of NSD2 was significantly correlated with lymph node metastasis,histological differentiation and TNM stage of breast cancer(respectively,χ^(2)=7.206,5.793,4.995,P<0.05).Conclusion Abnormal high expression of MDM2 and NSD2 proteins may be involved in the development and metastasis of breast cancer,and detection of MDM2 and NSD2 has certain reference value for evaluating the malignant degree of breast cancer and judging the metastatic potential of breast cancer.
