摘要
目的:探讨微小RNA(miR)-146a及白细胞介素-1受体相关激酶(IRAK1)在脑膜瘤组织中的表达及其与脑膜瘤临床特征的相关性。方法:选取50例脑膜瘤组织与10例因去骨瓣减压术获得的硬脑膜组织作为正常对照组,实时定量反转录聚合酶链反应(RT-qPCR)检测各组组织中miR-146a的表达水平及IRAK1 mRNA的相对表达水平;免疫组织化学方法检测IRAK1蛋白的表达水平;分析两者表达的相关性及其与脑膜瘤患者临床特征之间的关系。结果:脑膜瘤组织中miR-146a mRNA的表达水平明显低于正常脑膜组,IRAK1的表达水平明显高于正常脑膜组,且两者间的表达呈负相关(r=-0.305),差异均有统计学意义(t=-3.896、4.468,P值均<0.05)。WHOⅡ级、Ⅲ级的脑膜瘤组织中,miR-146a mRNA的表达水平均低于WHOⅠ级组(t=4.728、8.227),IRAK1 mRNA的表达水平均高于WHOⅠ级组(t=-4.262,t=-4.287),差异有统计学意义(P<0.05)。而WHOⅡ级与Ⅲ级间差异无统计学意义(P>0.05)。IRAK1蛋白的表达随着脑膜瘤组织各病理级别间差异明显。结论:miR-146a可能为脑膜瘤的抑癌基因,负调控IRAK1的表达。
Objective To explore the expression of microRNA(miR)-146a and interleukin-1 receptor associated kinases(IRAK1)in meningioma and their correlation with the clinical features of meningioma.Methods Totally,50 cases of meningioma tissues and 10 cases of dural tissues obtained after decompressive craniectomy were studied.MiR-146a and IRAK1 mRNA expression levels were detected by RT-PCR from different groups.The expression of IRAK1 protein was analyzed by immunohistochemistry.Pearson correlation analyses were performed to explore associations between miR-146a and IRAK1 mRNA expression.Also,the relationships between these changes with clinical data of the meningioma patients were analyzed.Results The expression level of miR-146a mRNA in meningioma tissue was significantly lower than that of the normal meningeal group,and the expression level of IRAK1 was significantly higher than that of the normal dura mater group,and the relationship between the two was negatively correlated(r=-0.305),the differences is statistically significant(t=-3.896,4.468,P<0.05).In WHO gradeⅡand gradeⅢmeningioma,the expression level of miR-146a mRNA was lower than the WHO gradeⅠgroup(t=4.728,8.227),and the expression level of IRAK1 mRNA was higher than the WHO gradeⅠgroup(t=-4.262,-4.287),the difference was statistically significant(P<0.05).But there was no statistically significant difference between WHO gradeⅡandⅢmeningioma(P>0.05).The expression of IRAK1 protein varies significantly with the pathological grades of meningioma(χ^(2)=20.044,P<0.05).Conclusion MiR-146a is a tumor suppressor gene for meningioma,and negatively regulates the expression of IRAK1.
