基于PacBio的三代测序的三七基因组测序分析 被引量:3

Genomic sequencing analysis of Panax notoginseng based on PacBio’s third-generation sequencing technology

摘要 目的丰富三七的基因组信息,为三七的开发利用提供依据。方法通过采摘新鲜的3年生三七叶,利用PacBio Sequel平台进行三七的基因组测序。结果在PacBio Sequel平台测得读取聚合酶729094个,总数据共25.80 GB。通过NT和NR注释,发现三七基因与胡萝卜、葡萄、芝麻、咖啡的相似比分别为:54.77%、6.22%、2.21%、2.18%。KOG发现一般功能预测所占基因数最多,达6545;其次为信号转导基因,达4437。KEGG发现三七中参与代谢的基因最多,达14064。通过SwissPort注释发现NR所特有的基因最多,达1279;KOG最少,仅为3。结论通过对三七叶的三代测序,有利于对三七进一步开发与利用,为三七的基因组研究奠定了基础,也为中药材现代化提供一定的帮助。 Objective Panax notoginseng as a valuable Chinese medicinal material, in order to enrich the genomic information of P. notoginseng, and to provide basis for the development and utilization of P. notoginseng.Methods To obtain the gene sequence of P. notoginseng,the study was conducted by picking fresh 3-year-old P. notoginseng leaves, and then to run sequenced by the PacBio Sequel platform. Results Three-generation sequencing of P. notoginseng leaves were used to read 729094 polymerases on PacBio Sequel platform, with a total data of 25.80 GB. Through NT and NR annotation, it was found that P. notoginseng gene had similar ratio with daucus carota, vitis vinifera, sesamum indicum, coffea canephora, respectively: 54.77%,6.22%,2.21%,2.18%. KOG found that general function predicted only is the most genes arrive at 6545.The second is signal transduction mechanisms gene, up to 4437. KEGG found that the leaf of P. notoginseng had the most genes involved in metabolism arrive at 14064. The gene specific to NR was found to be the most by SwissPort annotation arrive at 1279,the KOG is least, just for 3.Conclusion The three-generation sequencing of P. notoginseng leaf is conducive to its further development and utilization, which lays a foundation for the genomic research of P. notoginseng and also provides some help for the modernization of Chinese medicinal materials.
