摘要 目的:探讨并研究紫草中羟基萘醌总色素含量测定的稳定性。方法:以紫草为试材,参照2020版《中国药典》紫草项下羟基萘醌总色素含量的测定方法,测定紫草中羟基萘醌总色素的含量,对测定过程中的各影响因素进行逐一分析,总结影响紫草中羟基萘醌总色素含量测定稳定性的因素,并通过L9(34)正交试验优选出测定紫草中羟基萘醌总色素含量的最佳条件。结果与结论:在紫草药材粉碎成极细粉的条件下测得羟基萘醌总色素的含量值较为稳定。药粉细度是影响紫草中羟基萘醌总色素含量测定的主要因素,其次为药粉干燥时间及药粉含水量。将药材粉碎成极细粉、在50℃下干燥3 h、将药粉的含水量控制在9.0%左右是测定紫草中羟基萘醌总色素含量的最佳条件。 Objective:To explore and study the stability of the determination of total pigment content of hydroxynaphthoquinone in Persia sinensis.Methods:Taking Persia sinensis as the test material and referring to the determination method of total pigment content of hydroxynaphthoquinone under Persia sinensis in 2020 edition of Chinese Pharmacopoeia,the content of total pigment of hydroxynaphthoquinone in Persia sinensis was determined.The influencing factors in the determination process were analyzed one by one,and the factors affecting the stability of the determination of total pigment content of hydroxynaphthoquinone in Persia sinensis were summarized.L9(34)orthogonal test was used to determine the optimal conditions for the determination of total pigment content of hydroxynaphthoquinone in C.sinensis.RESULTS&CONCLUSION:The content of total pigment of hydroxynaphthoquinone was stable under the condition that the herb was crushed into very fine powder.The fineness of powder was the main factor affecting the determination of total pigment content of hydroxynaphthoquinone,followed by drying time and water content of powder.The best conditions for the determination of the total pigment content of hydroxynaphthoquinone in C.sinensis were to crush the medicinal material into very fine powder,dry it at 50℃for 3 h,and control the water content of the powder at about 9.0%.
