当归补血汤煎剂对骨髓抑制小鼠EPO、TPO、GM-CSF表达的影响研究

Effect of Danggui Buxue Decoction(当归补血汤煎剂) on Expressions of EPO,TPO and GM-CSF in Bone Marrow Suppressor Mice

目的探讨当归补血汤煎剂对骨髓抑制小鼠EPO、TPO、GM-CSF表达的影响研究。方法选取BALB/C纯系雄性小鼠100例,将其分为正常组、模型组、低剂量组、中剂量组以及高剂量组各20例。结果模型组骨髓抑制小鼠的层粘连蛋白(LN)以及纤维连接蛋白(FN)表达水平明显低于正常组(P<0.05),且中、高剂量组的LN以及FN表达水平明显高于低剂量组(P<0.05);模型组骨髓基质中的EPO、TPO以及GM-CSF等细胞因子表达水平明显高于正常组(P<0.05),且中、高剂量组的EPO、TPO以及GM-CSF等细胞因子表达水平改善情况明显优于低剂量组(P<0.05);中、高剂量组WBC、RBC、Hb、PLT以及BMC数量改善情况明显优于低剂量组(P<0.05);模型组骨髓基质中的MMP-2以及MMP-7蛋白表达水平明显高于正常组(P<0.05),IL-10水平低于正常组(P<0.05),且中、高剂量组MMP-2以及MMP-7蛋白表达水平明显低于低剂量组(P<0.05)。结论当归补血汤煎剂的在临床中的使用可有效促进骨髓抑制小鼠造血功能改善,有利于骨髓造血细胞实现快速增值,可推广使用。

Objective To investigate the effect of Danggui Buxue Decoction(当归补血汤煎剂) on the expressions of EPO, TPO and GM-CSF in bone marrow inhibited mice. Methods A total of 100 BALB/C pure male mice were selected and divided into normal group, model group, low dose group, medium dose group and high dose group, 20 cases in each. Results The expression levels of LN and FN in the model group were significantly lower than those in the normal group(P<0.05), and the expression levels of LN and FN in the medium and high dose groups were significantly higher than that in the low dose group(P<0.05). The expression levels of cytokines such as EPO, TPO and GM-CSF in the model group were significantly higher than those in the normal group(P<0.05), and the expression levels of cytokines such as EPO, TPO and GM-CSF in the medium and high dose groups were significantly better than those in the low dose group(P<0.05). The improvement of WBC, RBC, Hb, PLT and BMC in the medium and high dose groups was significantly better than that in the low dose group(P<0.05). The expression levels of MMP-2 and MMP-7 proteins in the bone marrow matrix of the model group were significantly higher than that of the normal group(P<0.05), and the levels of IL-10 were lower than that of the normal group(P<0.05), and the expression levels of MMP-2 and MMP-7 proteins in the medium and high dose groups were significantly lower than that of the low dose group(P<0.05). Conclusion The clinical application of Danggui Buxue Decoction can effectively promote the improvement of hematopoietic function in bone marrow inhibiting mice, and is conducive to the rapid proliferation of bone marrow hematopoietic cells.
