附子肉桂仙茅对不同状态大鼠免疫功能和能量代谢的影响 被引量:3

Effects of Fuzi(Aconm Lateralis Radix Praeparaia),Rougui(Cinnamomi Cortex)and Xianmao(Curculiginis Rhizoma)on Immune Function and Energy Metabolism of Rats in Different State

摘要 目的探讨辛热药附子、肉桂和仙茅对正常和虚寒状态大鼠免疫功能和能量代谢的影响。方法大鼠随机分为8组,即正常组、正常+附子组/肉桂组/仙茅组、虚寒组、虚寒+附子组/肉桂组/仙茅组,以注射氢化可的松琥珀酸钠建立虚寒状态模型,检测免疫功能指标(外周血白细胞数、胸腺脾脏指数、IL-4和IFN-γ含量)、能量代谢指标(肝和肌肉中SDH、ATP酶活性)。结果与正常组比较,虚寒组大鼠胸腺指数明显降低(P<0.05)、外周血白细胞数明显降低(P<0.001),血清中IFN-γ含量明显升高,IL-4含量降低,IFN-γ/IL-4比值升高(P<0.05),肝组织和肌肉组织中SDH的活性明显降低(P<0.05);肝组织和肌肉组织中Na^(+)-K^(+)-ATP酶,Mg^(2+)-ATP酶,Ca^(2+)-Mg^(2+)-ATP酶活性明显降低(P<0.05)。与虚寒组比较,虚寒+附子/肉桂/仙茅组白细胞数均显著升高(P<0.05),虚寒+附子组大鼠血浆IFN-γ含量显著降低(P<0.05);虚寒+肉桂组大鼠血浆IL-4含量显著升高(P<0.05),IFN-γ/IL-4比值显著降低(P<0.05);虚寒+仙茅组大鼠血浆IL-4含量显著升高(P<0.05)。肝组织中虚寒+仙茅组SDH活性均明显升高(P<0.05),虚寒+附子组Na^(+)-K^(+)-ATP酶,Mg^(2+)-ATP酶活性升高有统计学意义(P<0.05)。肌肉组织中,虚寒+附子/肉桂/仙茅组肌肉匀浆SDH活性均明显升高(P<0.05)。虚寒+附子/肉桂组Na^(+)-K^(+)-ATP酶,Mg^(2+)-ATP酶,Ca^(2+)-ATP酶活性升高(P<0.05)。结论附子肉桂仙茅能明显改善虚寒状态免疫功能低下和能量代谢障碍,对正常状态影响不明显。 Objective To investigate the effects of Fuzi(Aconm Lateralis Radix Praeparaia),Rougui(Cinnamomi Cortex)and Xianmao(Curculiginis Rhizoma)on immune function and energy metabolism in normal and deficiency cold rats.Methods Rats were randomly divided into 8 groups:normal group,normal+Fuzi(Aconm Lateralis Radix Praeparaia)group/Rougui(Cinnamomi Cortex)group/Xianmao(Curculiginis Rhizoma)group,deficiency cold group,deficiency cold+Fuzi(Aconm Lateralis Radix Praeparaia)group/Rougui(Cinnamomi Cortex)group/Xianmao(Curculiginis Rhizoma)group.The deficiency cold model was established by injecting hydrocortisone sodium succinate,and the immune function indexes(white blood cell count in peripheral blood,thymus spleen index,IL-4 and IFN-gamma content)were detected.Energy metabolism indicators(SDH,ATPase activity in liver and muscle)were observed.Results Compared with the normal group,the thymus index of rats in deficiency cold group was decreased significantly(P<0.05).The number of white blood cells in peripheral blood was decreased significantly(P<0.001).The content of IFN-gamma in serum was increased significantly,the content of IL-4 was decreased and the ratio of IFN-gamma to IL-4 was significantly(P<0.05).The activity of SDH in liver and muscle tissue of deficiency cold group was decreased significantly(P<0.05).The activities of Na^(+)-K^(+)-ATPase,Mg^(2+)-ATPase and Ca^(2+)-Mg^(2+)-ATPase in liver and muscle tissue were decreased significantly(P<0.05).Compared with those of the deficiency cold group,the number of white blood cells in deficiency cold+Fuzi(Aconm Lateralis Radix Praeparaia)/Rougui(Cinnamomi Cortex)/Xianmao(Curculiginis Rhizoma)group was increased significantly(P<0.05).The content of plasma IFN-gamma in the deficiency cold+Fuzi(Aconm Lateralis Radix Praeparaia)group was decreased significantly(P<0.05).The content of plasma IL-4 in deficiency cold+Rougui(Cinnamomi Cortex)group was increased significantly(P<0.05),and the ratio of IFN-gamma to IL-4 was decreased significantly(P<0.05).The content of IL-4 in plasma of rats in deficiency cold+Xianmao(Curculiginis Rhizoma)group was increased significantly(P<0.05).The activity of SDH in liver tissue of deficiency cold+Xianmao(Curculiginis Rhizoma)group was increased significantly(P<0.05),and that of the deficiency cold+Fuzi(Aconm Lateralis Radix Praeparaia)group was increased significantly(P<0.05).The activities of Na^(+)-K^(+)-ATPase and Mg^(2+)-ATPase in the subgroup were increased significantly(P<0.05).In muscle tissue,SDH activity of muscle homogenate in deficiency cold+Fuzi(Aconm Lateralis Radix Praeparaia)/Rougui(Cinnamomi Cortex)/Xianmao(Curculiginis Rhizoma)was group increased significantly(P<0.05).The activities of Na^(+)-K^(+)-ATPase,Mg^(2+)-ATPase and Ca^(2+)-ATPase in the deficiency cold+Fuzi(Aconm Lateralis Radix Praeparaia)/Xianmao(Curculiginis Rhizoma)group were increased(P<0.05).Conclusion Fuzi(Aconm Lateralis Radix Praeparaia),Rougui(Cinnamomi Cortex)and Xianmao(Curculiginis Rhizoma)can significantly improve the immune function and energy metabolism disorder in deficiency cold state,but have no obvious effect on normal state.
