摘要
目的筛选膝骨关节炎大鼠血清中疾病相关差异性代谢物,观察"易层"贴敷对膝骨关节炎大鼠代谢物的调控作用,探讨其治疗膝骨关节炎的作用机制。方法将18只SD大鼠随机均分为空白对照组、模型组和易层组,每组6只,通过切断前交叉韧带手术造模,造模成功后易层组连续外敷"易层"贴敷28 d,末次给药后处死大鼠,通过腹主动脉采血并收集各组滑膜组织。采用基于GC-MS的代谢组学技术分析各组大鼠血清样本中代谢物的变化,综合P<0.05、Fold Change>1.2条件筛选膝骨关节炎的疾病生物标志物,通过代谢通路富集分析初步探讨"易层"贴敷治疗膝骨关节炎的作用机制。结果膝骨关节炎大鼠滑膜出现炎性细胞浸润,多种代谢物代谢紊乱,"易层"贴敷能改善滑膜炎症状,并对多种代谢物起到回调作用。结论 "易层"贴敷主要通过调节机体内13个代谢物及4条相关代谢通路发挥治疗膝关节炎的作用。
Objective To screen the disease-related differential metabolites in serum of knee osteoarthritis(KOA) rats, observe the regulatory effect of layers adjusting application on the metabolites of knee osteoarthritis rats and explore the mechanism of its treatment of knee osteoarthritis. Methods SPF SD 18 rats were randomly divided into control group, model group and layers adjusting application group equally, 6 cases in each group. The KOA model was set up by cutting off the anterior cruciate ligament. After the model was successfully constructed, the layers adjusting application group applied layers adjusting application for 28 days continuously. After the final administration, blood and synovial tissues of each group were extracted. Metabolomics technology based on GC-MS was used to analyze the changes of metabolites in serum samples of rats in each group. Based on P<0.05 and Fold Change>1.2, disease biomarkers of KOA were screened and the action mechanism of layers adjusting application in the treatment of knee osteoarthritis was preliminarily explored through the enrichment analysis of metabolic pathways. Results The synovial tissues of KOA rats showed inflammatory cell infiltration and metabolic disorder of various metabolites. layers adjusting application can improve the inflammation of synovial tissues and play a role of callback to a variety of metabolites. Conclusion layers adjusting application is mainly applied by regulating 13 metabolites and 4 related metabolic pathways play a key role in the treatment of knee arthritis.
