基于网络药理学探讨玫芦消痤膏治疗痤疮的作用机制 被引量:3

摘要 目的采用网络药理学分析方法研究玫芦消痤膏(MXO)治疗痤疮(Acne)的作用靶点及信号通路,探索其作用机制。方法该研究利用中药系统药理学数据库和分析平台(TCMSP)筛选MXO活性成分和中药靶标,然后使用Uni Prot数据库进行基因名注释,并在Gene Cards数据库中检索痤疮的相关疾病靶标。利用Rversion 3.6.2软件、Cytoscape 3.7.1软件,构建中药-活性成分-靶点多维网络及MXO-Acne互作网络,利用STRING数据库,评估和整合蛋白之间的相互作用。最后利用R语言中Bioc Manager包进行基因本体功能注释及KEGG通路富集分析。结果共筛选得到MXO活性成分35个,槲皮素、山柰酚、木犀草素、花生四烯酸、β-胡萝卜素、芒柄花黄素、没食子酸和异鼠李素等是关键活性成分;作用靶标基因92个,核心蛋白有Rac-α丝氨酸/苏氨酸-蛋白激酶(AKT1)、白细胞介素(IL)-6、血管内皮生长因子A(VEGFA)、前列腺素内过氧化物合成酶2(PTGS2)和粒细胞趋化蛋白-1(CXCL8)等,主要涉及核受体活性、转录因子活性、类固醇激素受体活性、细胞因子活性等生物过程,KEGG通路主要富集在TNF、磷脂酰肌醇-3-激酶/蛋白激酶B(PI3K/Akt)、IL-17、AGE-RAGE等信号通路。结论预测了MXO治疗痤疮的潜在作用机制,为进一步的活性成分研究和实验验证提供了理论依据。 Objective To use the network pharmacology analysis method to study the effect target and signal pathway of Meilu Xiaocuo Ointment(MXO)in the treatment of acne(Acne),and to explore its action mechanism.Methods The study used the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP)to screen the MXO active components and TCM targets,and then the gene names annotation was performed by using the Uni Prot Database and the acne related disease targets were retrieved in the Gene Cards Database.The Rversion 3.6.2 software and Cytoscape 3.7.1 software were used to construct the multi-dimensional network of TCM-active component-target and MXO-Acne interaction network,and the STRING database was used to evaluate and integrate the interaction between proteins.Finally,the Bioc Manager package in the R language was used for conducting the gene ontology function annotation and KEGG pathway enrichment analysis.Results A total of 35 active components of MXO were screened out,quercetin,kaempferol,luteolin,arachidonic acid,β-carotene,formononetin,gallic acid and isorhamnetin were the key active components.There were 92 target genes,the core proteins included AKT1,IL-6,VEGFA,PTGS2,CXCL8,etc.,which were mainly involved in the nuclear receptor activity,transcription factor activity,steroid hormone receptor activity,cytokine activity and other biological processes.The KEGG pathway was mainly enriched in the signal pathways such as TNF,PI3K/Akt,IL-17 and age-rage.Conclusion This paper predicts the potential mechanism of MXO in treating acne,which provides the theoretical basis for further study and experimental verification of active ingredients.
