乳腺宁贴片外敷对乳腺增生模型大鼠乳腺组织的影响 被引量:3

摘要 目的观察外用药乳腺宁贴片对乳腺增生模型SD大鼠的乳头直径与高度、乳腺组织病理以及性激素受体表达的影响。方法将30只SD大鼠随机分为对照组(C组)、模型组(M组)和乳腺宁贴片组(T组),每组10只。M组与T组大鼠通过肌内注射苯甲酸雌二醇序贯黄体酮构建乳腺增生模型。从肌内注射开始,T组在大鼠第2对和第3对乳头外敷乳腺宁贴片中硫酸盐粉末膏药3 g,而C组和M组给予等量赋形剂,每24 h换药1次。干预第30天测量3组大鼠乳房的直径和高度,观察3组大鼠乳腺组织的病理学表现,运用免疫组化法检测各组大鼠雌激素受体α(ERα)、孕激素受体(PR)和Ki-67的表达情况。结果C、M、T组大鼠乳头直径分别为(1.40±0.07)、(1.64±0.05)、(1.51±0.03)mm,高度分别为(1.24±0.09)、(2.24±0.12)、(1.87±0.10)mm,与C组比较,M组大鼠的乳头直径、高度均显著增加(P<0.05或P<0.01);与M组比较,T组大鼠的乳头直径、高度均显著降低(P<0.05)。光镜下,与C组比较,M组乳腺小叶的腺泡数量显著增多,乳腺腺腔扩张同时出现大量分泌物,导管上皮细胞出现明显增生排列紊乱,ERα、PR和Ki-67表达均显著增加(P<0.01);与M组比较,T组乳腺小叶的腺泡数显著减少,管腔内的分泌现象减弱,ERα、PR和Ki-67表达均显著降低(P<0.05)。结论乳腺宁贴片外敷于乳腺增生模型SD大鼠,可降低乳腺组织ERα、PR和Ki-67表达,改善大鼠乳腺增生症。 Objective To observe the effects of Ruxianning plaster on diameter and height of nipple,histopathology of mammary gland and expression of sex hormone receptors in SD rats with mammary gland hyperplasia.Methods Thirty rats were randomly divided into control group(group C),model group(group M)and Ruxianning plaster group(group T),with 10 rats in each group.The groups M and T were given intramuscular injection of estradiol benzoate and progesterone to induce mammary gland hyperplasia.From the beginning of intramuscular injection,the rats in group T were externally treated with 3 g sulfate powder from Ruxianning plaster on the second and third nipples,while the rats in groups C and M were given the same amount of excipient.The dressing was changed every 24 hours.On the 30th day of intervention,the breast diameter and height were measured and the pathological manifestations were observed in all the three groups.In addition,the expression of estrogen receptor-α(ERα),progesterone receptor(PR)and Ki-67 was detected by immunohistochemistry.Results Compared with group C,the nipple diameter and height significantly increased in group M[(1.40±0.07)mm vs(1.64±0.05)mm and(1.24±0.09)mm vs(2.24±0.12)mm,respectively;P<0.05 or P<0.01].Compared with group M,the nipple diameter and height remarkably decreased in group T[(1.51±0.03)and(1.87±0.10)mm,respectively;P<0.05].Furthermore,compared with group C,increased number of acini in mammary lobule,dilated mammary gland cavity with abundant secretions,hyperplastic ductal epithelial cells,disordered arrangement,and up-regulated ERα,PR and Ki-67 expression were found in group M(P<0.01).However,compared with group M,the number of acini,mammary gland secretions and expression of ERα,PR and Ki-67 were decreased in group T(P<0.05).Conclusion Ruxianning plaster can down-regulate ERα,PR and Ki-67 expression and improve mammary gland hyperplasia in rats.
