原因不明复发性流产与先兆流产患者外周血单核细胞上Tim-3、PD-1与Th1/Th2细胞因子的关系 被引量:10

摘要 目的:探讨原因不明复发性流产(URSA)与先兆流产(TA)患者外周血单核细胞上T细胞免疫球蛋白黏蛋白-3(Tim-3)、程序性死亡因子-1(PD-1)与Th1/Th2细胞因子之间的关系。方法:选取2017年1月-2018年1月本院收治的URSA早孕患者40例为URSA组,40例早期先兆流产者为TA组,40例健康早孕孕妇为健康早孕组。测定外周血单核细胞上的Tim-3、PD-1表达水平,Th1/Th2细胞因子白介素-2(IL-2)、IL-12、γ-干扰素(IFN-γ)、IL-4、IL-6、IL-10。结果:URSA组和TA组的单核细胞上Tim-3、PD-1表达水平均低于健康早孕组且URSA组低于TA组,外周血IL-2、IL-12、IFN-γ水平均高于健康早孕组且URSA组高于TA组,外周血IL-4、IL-6、IL-10水平均低于健康早孕组且URSA组低于TA组(均P<0.05)。URSA组和TA组的外周血单核细胞上Tim-3与PD-1呈显著正相关性(P<0.05);Tim-3、PD-1表达与IL-2、IL-12、IFN-γ水平呈显著负相关性,而与IL-4、IL-6、IL-10水平呈显著正相关性(均P<0.05)。结论:外周血单核细胞上Tim-3、PD-1低表达可能参与了URSA和TA的发生发展,其机制可能与调节Th1/Th2细胞因子表达有关。 Objective: To investigate the relation of the expression of T cell immunoglobulin-mucin-3(Tim-3) and programmed death factor-1(PD-1) on monocytes in peripheral blood of women with unexplained recurrent spontaneous abortion(URSA) and threatened abortion(TA) to their Th1/Th2 cytokine level. Methods: From January 2017 to January 2018, 40 women with URSA during the first trimester of pregnancy were enrolled in group A, 40 women with TA during the first trimester of pregnancy were enrolled in group B, and 40 normal pregnant women were enrolled in group C. The Tim-3, PD-1 expression on monocytes, and the Th1/Th2 cytokines(IL-2, IL-12, IFN-γ, IL-4, IL-6, IL-10) level in peripheral blood of the women in the three groups were detected. Results: The expression levels of Tim-3 and PD-1 on monocytes of the women in group A and B were significant lower than those of the women in group C, and which of the women in group A were significant lower than that of the women in group B. The levels of IL-2, IL-12 and IFN-γof the women in group A and B were significant lower than those of the women in group C, and which of the women in group A were significant lower than that of the women in group B(all P<0.05). The expression level of Tim-3 on monocytes of the women in group A and B was significant positive correlation with their PD-1 expression level on monocytes in peripheral blood(P<0.05). The expression levels of Tim-3 and PD-1 were significant negative correlated with the levels of IL-2, IL-12, and IFN-γ, while which were significant positive correlated with the levels of IL-4, IL-6, and IL-10(all P<0.05). Conclusion: The low expression of Tim-3 and PD-1 on monocytes in peripheral blood of the pregnant women may be involved in their occurrence and development of URSA and TA, which mechanism may regulate the Th1/Th2 cytokine expression.
