摘要
目的:观察太芪培元颗粒对免疫低下小鼠外周血T细胞亚群含量、自然杀伤细胞(NK)计数及巨噬细胞吞噬作用的影响,初步探索该药可能的作用机制。方法:将BALB/c小鼠120只随机分为空白组、模型组、黄芪颗粒组、太芪培元颗粒低剂量组、中剂量组、高剂量组,除空白组外,其余各组均采用环磷酰胺注射法制备免疫低下小鼠模型,造模成功后分别灌服太芪培元颗粒及黄芪颗粒,4周后采血。结果:经造模处理后,模型组小鼠白细胞、淋巴细胞、胸腺系数和脾脏系数均低于正常组,差异有统计学意义(P<0.05),说明造模成功。经灌药处理后,太芪培元颗粒各治疗组NK细胞、白细胞、中性粒细胞数量均高于模型组,太芪培元颗粒高剂量组较模型组CD8^(+)细胞计数降低、CD4^(+)/CD8^(+)比值升高,太芪培元颗粒中剂量组与高剂量组淋巴细胞及单核细胞数量均高于模型组。结论:太芪培元颗粒可提高免疫低下小鼠白细胞、中性粒细胞、NK细胞数量,提示太芪培元颗粒对小鼠免疫功能的调节作用可能通过提高NK细胞数量实现。
Objective:To observe the effect of Taiqi peiyuan granule on T cell subpopulation,NK count and macrophage phagocytosis in peripheral blood of immunosuppressed mice,and to explore the possible mechanism of action.Methods:120 BALB/C mice were randomly divided into blank group,model group,Huangqi granules group,Taiqi peiyuan granules low dose group,medium dose group and high dose group.Except for the blank group,all the other groups prepared immunodeficient mouse model by cyclophosphamide injection.After successful modeling,Taiqi peiyuan granules and Huangqi granules were administrated respectively,and blood was collected 4 weeks later.Results:After modeling,the leukocyte,lymphocyte,thymus coefficient and spleen coefficient of the model group were significantly lower than normal group(P<0.05),indicating the success of modeling.After taking traditional Chinese medicine,The number of NK cells,leukocytes and neutrophils in Taiqi peiyuan granules in each treatment group was higher than that in the model group.The CD8^(+)cell count of Taiqi peiyuan Granule high-dose group was lower than that of model group,and the ratio of CD4^(+)/CD8^(+)increased.The number of lymphocytes and monocytes in Taiqi peiyuan Granules middle-dose group and high-dose group were higher than that in model group.Conclusion:Taiqi peiyuan Granules can increase the number of white blood cells,neutrophils,and NK cells in immunocompromised mice,suggesting that Taiqi peiyuan Granules may regulate the immune function of mice by increasing the number of NK cells.
