LINC01088对卵巢癌SKOV3细胞增殖、迁移与侵袭的调控作用及机制 被引量:2

摘要 目的:探讨长基因间非编码RNA 1088(LINC01088)对人卵巢癌SKOV3细胞增殖、迁移与侵袭的调控作用及可能机制。方法:携带LINC01088序列的重组慢病毒感染SKOV3细胞作为实验组(LV-LINC01088组),携带无意义序列的重组慢病毒感染SKOV3细胞作为阴性对照组(LV-NC组),同时将未感染的SKOV3细胞设为空白对照组(Control组)。CCK-8法和划痕实验分别用于检测细胞的增殖和迁移能力,Transwell实验检测细胞的迁移和侵袭能力。Western blot法分析上皮间质转化(EMT)相关蛋白、PI3K/Akt信号转导通路相关蛋白表达。结果:与Control组及LV-NC组比较,LV-LINC01088组SKOV3细胞的增殖、迁移与侵袭能力明显受到抑制(P<0.05),EMT相关蛋白E-cadherin表达升高,Vimentin表达降低,PI3K/Akt信号转导通路相关蛋白p-Akt 473、p-PI3K表达降低(P均<0.05)。结论:过表达LINC01088可抑制卵巢癌SKOV3细胞的增殖、迁移、侵袭及EMT,抑制PI3K/Akt信号转导通路可能是其作用机制。 Objective:To investigate the regulatory effect of long intergenic non-coding RNA 1088(LINC01088)on proliferation,migration and invasion of human ovarian cancer SKOV3 cells and its possible mechanism.Methods:The ovarian cancer cell line SKOV3 cells infected with recombinant lentivirus carrying LINC01088 sequence were used as the experimental group(LV-LINC01088 group)and infected with recombinant lentivirus carrying a nonsense sequence were used as the negative control group(LV-NC group),SKOV3 cells that not infected were set as the blank control group(Control group).CCK-8 method and scratch assay were used to detect the effect of LINC01088 on cell proliferation and migration ability.Transwell assay was used to detect the migration and invasion ability of cells.The protein expression levels of EMT related proteins and PI3K/Akt signaling pathway related proteins were analyzed using Western blot.Results:Compared with control group and LV-NC group,the proliferation、migration and invasion of SKOV3 cells were significantly inhibited in LV-LINC01088 group(P<0.05),the protein expression levels of EMT related proteins E-cadherin were up-regulated and the protein expression levels of Vimentin were down-regulated.PI3K/Akt signaling pathway related proteins p-Akt 473 and p-PI3K protein expressions were decreased in LV-LINC01088 group(P<0.05).Conclusions:Overexpression of LINC01088 can inhibit the proliferation、migration、invasion and EMT of SKOV3 cells which may relate to the inhibition of PI3K/Akt signaling pathway.
