摘要
目的探讨氧嗪酸钾联合不同造模剂对小鼠高尿酸血症(HUA)尿酸值的影响,为早期HUA的防治和药物筛选提供参考。方法采用300 mg/kg氧嗪酸钾、500 mg/kg尿酸、500 mg/kg次黄嘌呤、250 mg/kg乙胺丁醇多个造模剂,对ICR小鼠进行HUA造模,通过不同时间段取血测定尿酸值,比较不同造模因素对血尿酸的影响。结果给予300 mg/kg氧嗪酸钾能使小鼠尿酸水平升高,但是不能维持尿酸的高水平状态;给予300 mg/kg氧嗪酸钾联合500 mg/kg尿酸溶液能使血尿酸水平升高,且上升幅度较大(上升幅度为207.2%),但同样不能维持尿酸的高水平状态;给予300 mg/kg氧嗪酸钾联合500 mg/kg次黄嘌呤溶液能使血尿酸水平升高,且上升幅度较大(上升幅度为384.4%),但同样不能维持尿酸的高水平状态;给予300 mg/kg氧嗪酸钾联合250 mg/kg乙胺丁醇溶液能使血尿酸水平升高,上升幅度较大(上升幅度为320.8%),且能稳定维持尿酸在高水平的状态。结论氧嗪酸钾联合乙胺丁醇可成功使ICR小鼠尿酸值升高,并稳定维持尿酸的水平,造模效果优于单独的氧嗪酸钾、氧嗪酸钾联合尿酸、氧嗪酸钾联合次黄嘌呤。
Objective To explore the effect of potassium oxonate combined with different modeling agents on the uric acid value of mice with hyperuricemia(HUA),and provide reference for early prevention and treatment of HUA and drug screening.Methods HUA was modeled in ICR mice with 300 mg/kg potassium oxonate,500 mg/kg uric acid,500 mg/kg hypoxanthine and 250 mg/kg ethambutol.Blood samples were taken at different time periods to determine the uric acid value to compare the effects of different modeling factors on blood uric acid.Results The adminwastration of 300 mg/kg potassium oxonate could increase the level of uric acid,but could not maintain the high level of uric acid.The administration of 300 mg/kg potassium oxonate combined with 500 mg/kg uric acid solution could increase the level of uric acid in blood(the increase rate was 207.2%),but the high level of uric acid could not be maintained.The administration of 300 mg/kg potassium oxonate combined with 500 mg/kg hypoxanthine solution could increase the serum uric acid level(the increase rate was 384.4%),but the high level of uric acid could not be maintained either.The administration of 300 mg/kg potassium oxonate combined with 250 mg/kg ethambutol solution could increase the serum uric acid level(the increase rate was 320.8%),and the high level of uric acid could be maintained stably.Conclusion The combination of potassium oxazinate and ethambutol could successfully increase the uric acid value of ICR mice and stably maintain the level of uric acid.The modeling effect was better than that of single potassium oxonate,potassium oxonate combined with uric acid and oxygen,potassium oxonate combined with hypoxanthine.
