摘要
目的研究椿根皮Ailanthus altissima(Mill.)Swingle的化学成分及其体外抗肿瘤活性。方法椿根皮95%乙醇提取物采用硅胶、Sephadex LH⁃20进行分离纯化,根据理化性质及波谱数据鉴定所得化合物的结构。采用MTT法评价其体外抗肿瘤活性。结果从中分离得到15个化合物,分别鉴定为gigantol(1)、7α⁃H⁃9(10)⁃ene⁃11,12⁃epoxy⁃8⁃oxoeremophilane(2)、5,3′⁃dihydroxy⁃7,4′⁃dime⁃thoxyflavanone(3)、7β⁃H⁃9(10)⁃ene⁃11,12⁃epoxy⁃8⁃oxoeremophilane(4)、9⁃angeloyloxythymol(5)、对羟基苯乙酮(6)、coelonin(7)、crototropone(8)、7,8⁃dehydropenstemoside(9)、gentioside(10)、金圣草黄素(11)、4,5⁃二咖啡酰基奎宁酸甲酯(12)、monocerin(13)、isopropylidenkirenol(14)、1⁃(2⁃hydroxy⁃4⁃methylphe⁃nyl)propan⁃1,2⁃dione(15)。化合物8对AsPc细胞株、化合物13对COLO205和AsPc细胞株具有体外抑制作用。结论化合物1~2、4、8、13为首次从臭椿属植物中分离得到,所有化合物均为首次从椿根皮中分离得到,化合物8、13具有体外抗肿瘤活性。
AIM To study the chemical constituents from Ailanthus altissima(Mill.)Swingle and their antitumor activities in vitro.METHODS The 95%ethanol extract from A.altissima was isolated and purified by silica and Sephadex LH⁃20,then the structures of obtained compounds were identified by physicochemical properties and spectral data.Their antitumor activities in vitro were evaluated by MTT method.RESULTS Fifteen compounds were isolated and identified as gigantol(1),7α⁃H⁃9(10)⁃ene⁃11,12⁃epoxy⁃8⁃oxoeremophilane(2),5,3′⁃dihydroxy⁃7,4′⁃dime⁃thoxyflavanone(3),7β⁃H⁃9(10)⁃ene⁃11,12⁃epoxy⁃8⁃oxoeremophilane(4),9⁃angeloyloxythymol(5),p⁃hydroxyacetophenone(6),coelonin(7),crototropone(8),7,8⁃dehydropenstemoside(9),gentioside(10),chrysoeriol(11),4,5⁃di⁃caffeo ylquinate(12),monocerin(13),isopropylidenkirenol(14),1⁃(2⁃hydroxy⁃4⁃methylphe⁃nyl)propan⁃1,2⁃dione(15).Compound 8 inhibited AsPc cells,and compound 13 inhibited COLO205 and AsPc cells in vitro.CONCLUSION Compounds 1-2,4,8,13 are isolated from genus Ailanthus for the first time,and all the compounds are isolated from this plant for the first time,and compounds 8 and 13 have inhibitory activities in vitro.
