龙骨风HPLC指纹图谱建立及5种成分测定 被引量:6

摘要 目的建立不同产地龙骨风Alsophila spinulosa(Wall.ex Hook.)Tryon HPLC指纹图谱,并同时测定5种成分的含量。方法龙骨风甲醇⁃1%冰醋酸溶液的分析采用COSMOSIL 5C18⁃PAQ色谱柱(4.6 mm×250 mm,5μm);流动相甲醇⁃0.1%磷酸,梯度洗脱;体积流量1.0 mL/min;柱温35℃;检测波长280 nm。结果11批样品指纹图谱中有15个共有峰,相似度均大于0.829。原儿茶酸、原儿茶醛、咖啡酸、反式对羟基肉桂酸和阿魏酸分别在0.1879~27.0576μg(r=0.9999)、0.3710~13.3560μg(r=0.9998)、0.1704~12.0600μg(r=0.9998)、0.0235~6.0109μg(r=0.9999)、0.0198~1.2200μg(r=0.9999)范围内线性关系良好,平均加样回收率98.98%~102.62%,RSD<3.0%。结论该方法稳定可靠,可用于龙骨风的质量控制。 AIM To establish the HPLC fingerprints of Alsophila spinulosa(Wall.ex Hook.)Tryon.and to determine the contents of five constituents.METHODS The analysis of methyl alcohol⁃1%glacial acetic acid extract of A.spinulosa was developed on a 30℃thermostatic COSMOSIL 5C18⁃PAQ chromatographic column(250 mm×4.6 mm,5μm),with the mobile phase comprising of methyl alcohol⁃0.1%phosphoric acid flowing at 1.0 mL/min in a gradient elution manner,and the detection wavelength was set 280 nm.RESULTS There were fifteen common peaks in the fingerprints of eleven batches of samples,with the similarities of more than 0.829.Protocatechuic acid,protocatechuic aldehyde,caffeic acid,p⁃coumaric acid,and ferulic acid showed good linear relationships within 0.1879-27.0576μg(r=0.9999)、0.3710-13.3560μg(r=0.9998)、0.1704-12.0600μg(r=0.9998)、0.0235-6.0109μg(r=0.9999)、0.0198-1.2200μg(r=0.9999),whose average recoveries were 98.98%-102.62%with all RSDs less than 3.0%.CONCLUSION This stable and reliable method can be used for the quality control of A.spinulosa.
