乳腺癌外泌体中小分子RNA表达分析

Expression analysis of small RNA in breast cancer exosome

目的分析乳腺癌外泌体中小分子RNA(small RNA)的表达。方法采用NextSeq CN500测序仪检测乳腺癌细胞株外泌体和人正常乳腺上皮细胞株外泌体中small RNA的表达,DEGseq R包分析各small RNA表达差异,log2FC>1,FDR<0.05为差异有统计学意义。结果乳腺癌细胞株外泌体相对人正常乳腺上皮细胞株外泌体、34种微小RNA(miRNA)、39种转运RNA来源的小RNA(tsRNA)、和17种tsRNAs表达上调;104种miRNAs、21种与Piwi蛋白相作用的RNA和21种tsRNAs表达上调。结论乳腺癌细胞株外泌体与人正常乳腺上皮细胞株外泌体间small RNA存在表达差异。

Objective To analysis the expression of small RNA(small RNA)in breast cancer exosome.Methods Nextseq cn500 sequencer was used to detect the small RNA expression in the exosome of breast cancer cell line and the exosome of normal breast epithelial cell line.Degseq R package was used to analyze the differences of small RNA expression.Log2fc>1 and FDR<0.05 were considered statistically significant.Results Compared with the exosome of normal breast epithelial cell line,there were 34 miRNAs,39 piRNAs and 17 tsRNAs were up-regulated;104 miRNAs,21 piRNAs and 21 tsRNAs down-regulated in exosome of breast cancer cell line.Conclusion The expression of small RNA in exosome of breast cancer cell line was different from that of normal breast epithelial cell line.
