摘要
目的探讨Apelin-13对肝细胞癌HepG2细胞增殖的影响。方法将HepG2细胞按Apelin-13不同浓度分组:10%胎牛血清组(对照组)、0.0001、0.001、0.01、0.1μmol/L Apelin-13组,处理时间为24 h;按Apelin-13时效关系分组:0、6、12、18、24 h组,处理浓度为0.1μmol/L。细胞增殖实验检测各组处理后细胞水平。蛋白质印迹法检测细胞增殖相关蛋白细胞周期蛋白D1(CyclinD1)和细胞凋亡相关蛋白半胱氨酸天冬氨酸蛋白酶-3(Caspase-3)的表达水平。结果HepG2细胞增殖程度随Apelin-13浓度增加及作用时间延长而增强(P<0.05)。CyclinD1蛋白表达水平随Apelin-13处理浓度的升高及处理时间延长而升高(P<0.05);Caspase-3蛋白表达水平随Apelin-13处理浓度的升高及处理时间延长而降低(P<0.05)。结论Apelin-13在一定范围内促进肝细胞癌HepG2细胞增殖,抑制Caspase-3蛋白的表达,抑制效果随着处理时间延长和浓度的升高而增强。
Aim To investigate the effect of Apelin-13 on proliferation of HepG2 cells.Methods HepG2 cells were divided into different concentrations of Apelin-13:10%FBS(control),0.0001,0.001,0.01,0.1μmol/L.And the different durations were 0 h(control),6 h,12 h,18 h,24 h.The number of cells in each group was calculated by MTT method.Western blot was used to detect the expressions of cell cycle protein D1(Cyclin D1)and cysteine aspartate protease 3(Caspase-3).Results The proliferation of HepG2 cells was increased with the increase of Apelin-13 concentration,and the prolongation of the Apelin-13 treatment time(P<0.05).The expression of Cyclin D1 was increased with the increase of Apelin-13 concentration and treatment time,while the expression of Caspase-3 decreased with the increase of Apelin-13 concentration and treatment time.Conclusion Apelin-13 promoted the proliferation of hepatocellular carcinoma HepG2 cells and inhibited the expression of Caspase-3 protein within a certain range,and the inhibitory effect was enhanced with prolongation of treatment time and the increase of treatment concentration.
