Expression of TRIM38 and MITA in pregnant mice and their effects

目的:探讨TRIM38、MITA在小鼠妊娠过程中的表达及其意义。方法:构建正常组孕鼠及流产组小鼠模型,于孕11.5 d取蜕膜组织并提取巨噬细胞,RT-PCR及Western Blot检测TRIM38、MITA的含量;并证实TRIM38与MITA表达存在的相关性;且通过Vevo2100成像系统检测孕鼠子宫动脉血流,以找到TRIM38及MITA表达对妊娠的影响。结果:RT-PCR及Western Blot结果均显示流产小鼠蜕膜及巨噬细胞TRIM38、MITA的表达升高;且TRIM38与MITA的表达呈正相关;MITA高表达质粒转染后,小鼠流产率明显增加,且流产小鼠的子宫血流阻力指数明显升高,提示TRIM38及MITA对妊娠有一定的影响。结论:TRIM38可能通过调控巨噬细胞中MITA的表达引发流产,且可能和泛素化有关,其作用机制需近一步探讨,为临床上治疗该类患者提供新思路。

Objective:To investigate the expression and significance of TRIM38 and MITA during pregnancy in mice.Methods:The mice models of normal pregnancy and abortion were constructed.The decidual tissues were taken and the macrophages were extracted on day 11.5 of pregnancy.The contents of TRIM38 and MITA were detected by RT-PCR and Western Blot;and the correlation between TRIM38 and MITA expression was confirmed.The Vevo2100 imaging system was used to detect the blood flow of uterine arteries in pregnant rats to find the effects of TRIM38 and MITA expression on pregnancy.Results:Both RT-PCR and Western Blot results showed that the expression of TRIM38 and MITA both in decidua and macrophages of aborted mice increased.The expression of TRIM38 and MITA was positively correlated.After MITA expression plasmid transfection, the abortion rate of mice increased significantly, and the abortion mice’s uterine blood flow resistance index increased significantly, suggesting that TRIM38 and MITA have certain effects during pregnancy.Conclusion:TRIM38 may induce abortion by regulating the expression of MITA in macrophages, and may be related to ubiquitination.The specific mechanism needs to be further explored to provide new ideas for clinical treatment of this type of patients.
