电针结合康复训练对脑缺血小鼠的运动功能改善作用和分子机制研究

Improvement of motor function and molecular mechanism of electroacupuncture combined with rehabilitation training in mice with cerebral ischemia

摘要 目的探讨电针结合康复训练对脑缺血小鼠的运动功能改善作用和分子机制。方法以C57BL/6J小鼠为研究对象,随机分为对照组、假手术组,模型组以及电针结合康复训练组(治疗组),每组各10只小鼠,首先分别观察各组小鼠治疗前,治疗后1d、7d、14d的神经功能缺损评分和运动行为学评分,通过qPCR、Western blot检测小鼠脑组织RORA和NF-κB的表达水平变化。随后以RORA敲除的转基因小鼠为研究对象,再次构建脑缺血小鼠模型,观察电针结合康复训练对小鼠神经功能缺损评分和运动行为学评分的影响。结果模型组小鼠神经功能缺损评分和运动行为学评分均明显高于对照组和假手术组(P<0.05),经电针结合康复训练后7d和14d,治疗组上述评分均明显低于单纯造膜未治疗的小鼠(P<0.05)。训练14d后小鼠脑组织RORA的表达水平明显升高,而p-P65的表达水平明显降低(P<0.05)。进一步采用电针结合康复训练治疗RORA敲除的脑缺血损伤小鼠,发现治疗前和治疗后1d、7d、14d的神经功能和运动行为学评分均无明显差异(P>0.05)。结论电针结合康复训练可通过促进小鼠脑组织表达RORA,抑制NF-κB信号通路,从而促进脑缺血小鼠神经和运动功能的改善。 Objective To study the effect and molecular mechanism of electroacupuncture combined with rehabilitation training on motor function in mice with cerebral ischemia. Methods C57 BL/6 J mice were randomly divided into control group, sham operation group, model group and electroacupuncture combined with rehabilitation training group(treatment group), with 10 mice in each group. The expression levels of RORA and NF-κB signaling pathway were detected by western blot. Then, we used RORA knockout transgenic mice as the research object, and constructed the cerebral ischemia mouse model again, and observed the influence of electroacupuncture combined with rehabilitation training on the neurological deficit score and motor behavior score of mice. Results After modeling, the neurological deficit score and motor behavior score of the model group were significantly higher than those of the control group and the sham operation group(P<0.05). After 7 and 14 days of electroacupuncture combined with rehabilitation training, the above scores in the treatment group were significantly lower than those in the control group and the untreated group(P<0.05). After 14 days of training, the expression of RORA was significantly increased, while the expression of p-p65 was significantly decreased(P<0.05). Further, electroacupuncture combined with rehabilitation training was used to treat RORA knockout mice with cerebral ischemia. There was no significant difference in neurological function and motor behavior scores before and 1, 7 and 14 days after treatment(P>0.05). Conclusion Electroacupuncture combined with rehabilitation training can improve the neurological and motor function of cerebral ischemia mice by promoting the expression of RORA and inhibiting NF-κ B signaling pathway.
