Screening of diagnostic and prognostic biomarkers of gastric cancer based on bioinformatics analysis

目的本研究旨在通过生物信息学方法筛选新的胃癌诊断及预后标志物。方法从GEO数据库GSE54129和TCGA-STAD数据集中筛选重叠差异表达基因(differentially expressed genes, DEGs)。通过GO和KEGG通路分析,探讨这些基因的功能。利用蛋白质相互作用网络分析确认DEGs之间的枢纽基因。通过ROC曲线分析和Cox风险比分析,阐明枢纽基因在诊断和预后中的作用。结果本研究共筛选出222个重叠基因,富集于细胞外基质等相关通路。我们进一步确定了17个枢纽基因,其中BGN、COMP、COL5A2和SPARC可能是胃癌的重要诊断和预后指标。结论我们确定了BGN、COMP、COL5A2和SPARC四种潜在的胃癌生物标志物,可能作为胃癌的诊断和预后指标。

Objective We aimed to explore new diagnostic and prognostic indicators for gastric cancer(GC) in the current study. Methods We screened the overlapped differentially expressed genes(DEGs) from GSE54129 and TCGA STAD datasets. Protein-protein interaction network analysis recognized the hub genes among these DEGs. Gene Ontology and KEGG pathway analysis were processed to investigate the role of these genes. ROC curve analysis and Cox hazard ratio analysis were carried out to clarify the diagnostic and prognostic role of these genes. Results A total of 222 overlapping genes were screened, which enriched in extracellular matrix related pathways. Further, 17 hub genes were identified, and our findings demonstrated that BGN, COMP, COL5A2 and SPARC might be important diagnostic and prognostic indicators of GC. Conclusion We identifiedfour potential biomarkers of GC, including BGN, COMP, COL5A2 and SPARC, which have the ability tobe diagnosticand prognostic indicators for GC.
