獐牙菜苦苷治疗糖尿病周围神经痛的机制研究

Efficacy of Swertiamarin as a treatment of diabetic peripheral neuralgia:A mechanistic study

摘要 目的探索獐牙菜苦苷治疗糖尿病周围神经痛(DPN)大鼠的疗效。方法随机将32只SD大鼠(SPF级)分4组:空白对照组、DPN模型组、獐牙菜苦苷治疗组以及还原型烟酰胺腺嘌呤二核苷酸磷酸氧化酶(NOXs)抑制剂组。在造模成功后第1天(D1)、第7天(D7)、第14天(D14)用药后2 h分别检测各组大鼠血糖、热辐射缩足潜伏期、鼠尾感觉神经传导速度(SNCV)以及鼠尾血流灌注量测量。结果与空白对照组相比,DPN组大鼠血糖升高、热刺激潜伏期缩短、鼠尾的血流灌注量以及鼠尾SNCV下降(P<0.05)。相比之下,在经过獐牙菜苦苷或者NOXs抑制剂药物干预后,2组热刺激潜伏期明显延长(P<0.05),鼠尾的血流灌注量增加(P<0.05),鼠尾SNCV加快(P<0.05)。獐牙菜苦苷可以降低血糖(P<0.05),NOXs抑制剂并无调节血糖功能(P>0.05)。鼠尾SNCV在D1天,獐牙菜苦苷组优于NOXs抑制剂组。其余指标两个治疗组间无统计学差异(P>0.05)。结论獐牙菜苦苷可以调节血糖,提高痛域,增强肢体神经血液灌注,营养神经,提高SNCV治疗DPN。 Objective To investigate the therapeutic effect of Swertiamarin as a treatment of diabetic peripheral neuralgia(DPN)in rats.Methods Thirty-two specific pathogen-free SD rats were randomly divided into four groups:blank control group,DPN model group,Swertiamarin treatment group,and NOXS inhibitor treatment group.On days 1,7 and 14 after successful modeling of DPN,the rats were measured for blood glucose,thermal paw withdrawal latency,caudal sensory nerve conduction velocity(SNCV)and caudal blood perfusion.Results Compared with the blank control group,the rats in DPN model group had increased blood glucose,shortened thermal paw withdrawal latency,and decreased caudal blood perfusion and SNCV(P<0.05).In contrast,after intervention with Swertiamarin or NOXS inhibitor,the treatment groups showed significantly prolonged thermal paw withdrawal latency(P<0.05),increased caudal blood perfusion(P<0.05),and faster caudal SNCV(P<0.05).Swertiamarin was capable to lower the blood sugar(P<0.05),but such effect was not observed with NOXS inhibitor(P>0.05).On day 1,the caudal SNCV was faster in the Swertiamarin treatment group compared with the NOXS inhibitor treatment group.There were no statistical differences in other indicators between the two treatment groups(P>0.05).Conclusion Swertiamarin can regulate blood sugar,improve pain threshold,enhance blood perfusion and nutrition of limb nerves,increase SNCV,and ultimately ameliorate DPN.
