摘要
目的建立准确、高效、简便的反相高效液相色谱法(RP-HPLC)同时测定不同产地桑白皮药材中桑辛素M、桑黄酮G、桑黄酮C、桑皮酮E和桑辛素5种指标成分含量,为桑白皮药材质量评估提供依据。方法色谱柱为DikmaPlatisilTM ODS-C18柱(150 mm×4.6 mm,5μm),选择乙腈(含体积分数0.3%甲酸)-水(含体积分数0.3%甲酸)作为流动相,梯度洗脱;流速为1.0 mL·min-1;柱温为35℃;检测波长为270 nm。结果桑辛素M、桑黄酮G、桑黄酮C、桑皮酮E和桑辛素的质量浓度分别在2.0~40 mg·L^(-1),10.0~200 mg·L^(-1),10.0~200 mg·L^(-1),2.0~40 mg·L^(-1)和2.0~40 mg·L^(-1)内线性关系良好(r≥0.9991),平均加样回收率分别为94.6%,92.6%,93.5%,92.0%和92.6%,RSD分别为2.5%,2.3%,1.2%,1.4%和1.9%(n=6)。结论所建立的方法准确、重现性好,适用于桑白皮药材中桑辛素M、桑黄酮G、桑黄酮C、桑皮酮E和桑辛素的含量测定,为评价桑白皮的质量提供参考。
Objective To develop a simple,effective and accurate RP-HPLC method for simultaneous determination of moracin M,kuwanon G,kuwanon C,kuwanon E and morusin in Mori Cortex from different regions,which provided a basis for the evaluation and quality control of Mori Cortex.Methods The analysis method was carried out by high performance liquid chromatography.The chromatographic separation was performed on a Dikma PlatisilTM ODS-C18 column(150 mm×4.6 mm,5μm)using gradient elution with mobile phase consisting of 0.3%formic acid in both acetonitrile and water.The flow rate was 1.0 mL·min-1,the column temperature was 35℃,and the detection wavelength was 270 nm.Results Calibration curves were linear(r≥0.9991)in the range of 2.0-40 mg·L^(-1)for moracin M,10.0-200 mg·L^(-1)for kuwanon G,10.0-200 mg·L^(-1)for kuwanon C,2.0-40 mg·L^(-1)for kuwanon E and 2.0-40 mg·L^(-1)for morusin.The average recoveries of moracin M,kuwanon G,kuwanon C,kuwanon E and morusin were 94.6%,92.6%,93.5%,92.0%and 92.6%,while the relative standard deviations(RSDs)were 2.5%,2.3%,1.2%,1.4%and 1.9%(n=6),respectively.Conclusion The method is accurate and reliable,and can be applied successfully to simultaneous determination of morusin,moracin M,kuwanon G,kuwanon C,kuwanon E and morusin in Mori Cortex,which provides quality control of Mori Cortex.
