摘要
目的探讨石蒜碱对肠癌细胞生长和转移的影响。方法将HCT116和HT29细胞分为四组,分别使用质量分数0.9%NaCl溶液、10μmol·L^(-1)、20μmol·L^(-1)石蒜碱和5-氟尿嘧啶(5-FU)处理。之后通过MTT实验检测药物作用0、12、24、36和48 h时细胞的增殖情况,计算石蒜碱对细胞活力的抑制情况,膜联蛋白V-碘化丙啶检测细胞的凋亡情况,Transwell实验检测细胞的迁移状况。Western blot实验检测质量分数0.9%NaCl溶液、10μmol·L^(-1)和20μmol·L^(-1)石蒜碱对细胞中Cyclin D1、bcl-2和MMP2蛋白的影响。结果石蒜碱能够显著抑制HCT116细胞和HT29细胞的增殖。10μmol·L^(-1)或20μmol·L^(-1)的石蒜碱作用24 h时对HCT116细胞增殖的抑制率为(46.94±1.41)%和(58.48±3.04)%。10μmol·L^(-1)或20μmol·L^(-1)的石蒜碱作用24 h时对HT29细胞增殖的抑制率为(46.22±3.56)%和(56.44±2.99)%。石蒜碱作用24 h时对HCT116和HT29细胞的IC50为11.66μmol·L^(-1)和12.78μmol·L^(-1)。石蒜碱能够促进细胞的凋亡抑制细胞的迁移。10μmol·L^(-1)或20μmol·L^(-1)的石蒜碱作用24 h时能够显著下调Cyclin D1、bcl-2和MMP2蛋白的表达。结论石蒜碱能够抑制肠癌细胞的生长和迁移。
Objective To investigate the effect of lycorine on the growth and metastasis of colorectal cancer cells.Methods HCT116 and HT29 cells were divided into three groups and treated with NaCl,10μmol·L^(-1),20μmol·L^(-1)lycorine and 5-Fu respectively.MTT assay was used to detect cell proliferation at 0 h,12 h,24 h,36 h and 48 h,inhibition of lycorine on cell viability was calculated,apoptosis was detected by Annexin V-propidium iodide,and cell migration was detected by transwell assay.Western blot assay was used to detect the effects of NaCl,10μmol·L^(-1)and 20μmol·L^(-1)lycorine on Cyclin D1,Bcl-2 and MMP2 proteins in HCT116 cells.Results Lycorine could significantly inhibit the proliferation of HCT116 cells and HT29 cells.The inhibitory rates of 10μmol·L^(-1)and 20μmol·L^(-1)lycorine on the proliferation of HCT116 cells were(46.94±1.41)%and(58.48±3.04)%at 24 h.The inhibitory rates of 10μmol·L^(-1)and 20μmol·L^(-1)lycorine on the proliferation of HT29 cell were(46.22±3.56)%and(56.44±2.99)%after 24 h of lycorine treatment at 10μmol·L^(-1)and 20μmol·L^(-1).The IC50 of lycorine on HCT116 and HT29 cells was 11.66μmol·L^(-1)and 12.78μmol·L^(-1)for 24 h.Lycorine could promote cell apoptosis and inhibit cell migration.The expression of Cyclin D1,Bcl-2 and MMP2 protein was significantly down-regulated by lycorine at 10μmol·L^(-1)and 20μmol·L^(-1)for 24 h.Conclusion Lycorine can inhibit the growth and migration of colon cancer cells.
