五种处理对奶油草莓组培苗脱毒效果的影响 被引量:2

摘要 为筛选出奶油草莓组培苗的脱毒培养方法及病毒检测最佳方法,利用含有病毒的奶油草莓老叶作外植体,诱导出含病毒的试管苗,分别用含病毒唑的培养基、昼夜变温处理、高温培养、低温培养、茎尖脱毒培养五种方法进行脱毒处理,并对脱毒处理的试管苗分别采用:涂抹曼陀罗、苋色黎、千日红3种指示植物后隔离移栽,移栽后3周对其外部形态观察检测。结果表明:40℃的高温处理的茎尖脱毒效果最好,含三氮唑核苷25 mg/L的培养基有利于培养奶油草莓脱毒苗,指示植物曼陀罗对奶油草莓病毒比苋色黎、千日红更敏感,可作草莓病毒的指示植物。 To screen out the best method of detoxification culture and virus detection of tissue culture seedling of Strawberry of cream,the old leaves of strawberry with whipped cream are used as the explants to seed out of the tube seedlings and develop the tube seedlings respectively.Five methods-ribavirin in medium composition,day and night temperature treatment,high temperature,low temperature cultivation,seedling stem tip cultivation-are used to sterilize the tube seedlings,which are then treated with indicator plant daub mandala,amaranth,thousand-day red Chrysanthemum.Three weeks after transplanting,the external morphology was detection.The results showed that 1)the detoxification effect was the best after 40℃ high temperature treatment combined with stem tip culture;2)the medium containing 25 mg/L triazole riboside was beneficial to the detoxification of strawberry butters;and 3)the plant Mandala is more sensitive to the strawberry virus than amaranth and thousand-day red Chrysanthemum and is thus the most convenient method for observation of external morphology after transplanting of detoxified seedlings.
