摘要
为探究miR-15b在荣昌猪不同体组织中的表达差异及其在C_(2)C_(12)细胞不同发育时期的动态表达变化,本研究采用RT-qPCR方法检测miR-15b在荣昌猪心脏、肝脏、脾脏、肺、肾脏、胃、肌肉、胸腺和小肠组织及其在C_(2)C_(12)细胞增殖和分化过程中的表达水平。结果表明:miR-15b在荣昌猪各组织中均有表达,其中在心脏和胸腺中的表达量最高,在肝脏和肾脏中的表达量最低;miR-15b的表达量在C_(2)C_(12)细胞的整个增殖过程中均显著升高,在C_(2)C_(12)细胞的整个分化过程中先显著升高再显著降低。综上所述,miR-15b在荣昌猪各组织广泛表达,其表达量在C_(2)C_(12)细胞的增殖和早期分化中显著升高,推测miR-15b在C_(2)C_(12)细胞的增殖以及分化早期发挥重要功能。
In order to explore the expression characteristics of miR-15b in different tissues of pigs and different developmental stages of C_(2)C_(12) cells,RT-qPCR was used to detect the expression level of miR-15b in different tissues of Rongchang pigs and different stages of proliferation and differentiation of C_(2)C_(12) cells.The results showed that miR-15b is expressed in all tissues of Rongchang pigs,with higher expression level in heart and thymus and lower expression level in liver and kidney.The expression of miR-15b increased significantly during the proliferation process of C_(2)C_(12) cells and early differentiation of C_(2)C_(12) cells,but it’s reduced in the late differentiation of C_(2)C_(12) cells.In general,miR-15b is widely expressed in various tissues of Rongchang pigs.The expression of miR-15b increased significantly in the proliferation and early differentiation of C_(2)C_(12) cells,which suggested that miR-15b may play an important role in the proliferation and early differentiation of C_(2)C_(12) cells.
