稀释方法、孵育温度和时间对冷冻-解冻后猪精液质量的影响 被引量:6

摘要 试验以精子冻后活率、质膜完整率和顶体完整率三项指标评价了一步稀释法(Ⅰ组)和两步稀释法(Ⅱ组),以及不同保存温度和时间对解冻、稀释后的猪精液质量的影响。结果表明,采用两步稀释法对解冻后的猪细管冻精进行稀释,精子活率、质膜完整率和顶体完整率分别达到82.6%、89.4%和90.3%,极显著高于一步稀释法(P<0.01)。用两步法稀释后的精液36℃保存5 min、10 min和15 min时精子活率分别为81.2%、80.8%和80.9%(P>0.05),40 min时为50.1%;精子质膜完整率和顶体完整率未见显著下降(P>0.05);17℃保存15 h时精子活率、质膜完整率和顶体完整率分别为81.1%、88.9%和91.4%(P>0.05);60 h时分别为50.4%、61.4%和61.6%,均极显著低于初始的各项指标(P<0.01)。 Three indices,semen motility rate after freezing,plasma membrane integrity rate and acrosomal integrity were used to evaluate the effect of one-step diluting method(group I)and two-step diluting method(group II),different storage temperature and time on quality of thawed and diluted swine semen.The results showed with two-step diluting method,semen motility rate after freezing,plasma membrane integrity rate and acrosomal integrity were 82.6%,89.4%and 90.3%respectively,all significantly higher than that of one-step method(P<0.01).When the semen thawed using two-step diluting method were put into 36℃thermostat water bath and kept for 5 min,10 min and 15 min,the semen motility rate were 81.2%,80.8%and 80.9%respectively.Then the semen motility rate dropped rapidly to 50.1%at 40 min,but plasma membrane integrity rate and acrosomal integrity did not drop significantly during the whole storage(P>0.05).When the semen were stored into 17℃incubator,the semen motility rate maintained for a longer time.Till 15 h,the semen motility rate,plasma membrane integrity rate and acrosomal integrity were 81.1%,88.9%and 91.4%(P>0.05),respectively;Till 60 h,all three indexes dropped rapidly to 50.4%,61.4%and 61.6%,all significantly lower than initial indexes(P<0.01).
