摘要
目的本研究旨在探讨天灸药物白芥子散对哮喘大鼠免疫稳态重建及其对于RhoA基因表达的影响。方法60只Wistar大鼠分为6组,A组(正常对照组)、B组(阴性对照组)、C组(哮喘模型组)、D组(地塞米松组)、E组(白芥子散2 h组)以及F组(白芥子散4 h组),每组10只。造模成功后,A组、B组和C组正常饲养;D组腹腔注射地塞米松;E和F组背部剃毛,并在其双侧肺俞、脾俞、肾俞采用白芥子散分别艾灸2 h和4 h。检测和比较6组大鼠支气管肺泡灌洗液中的总细胞数、中性粒细胞数以及巨噬细胞数,白细胞介素-6和肿瘤坏死因子(TNF)-α的表达水平,以及肺组织中RhoA基因的蛋白质表达水平。结果肺组织苏木素-伊红(HE)染色结构显示,采用地塞米松和白芥子散治疗2 h和4 h后肺泡结构的破坏,炎性细胞的侵润明显减轻。与A组相比较,C组支气管肺泡灌洗液中的总细胞、中性粒细胞和巨噬细胞的数量均显著增加(均P<0.01),TNF-α和IL-6水平均显著增加(均P<0.01),肺组织中的RhoA基因的蛋白质表达水平显著增加。地塞米松和白芥子散2 h和4 h治疗显著减少了支气管肺泡灌洗液中的总细胞、中性粒细胞和巨噬细胞的数量(均P<0.01),TNF-α和IL-6水平均显著增加(均P<0.01),以及肺组织中的RhoA基因的蛋白质表达水平。结论天灸药物白芥子散可改善抗原诱导的大鼠哮喘,帮助其免疫稳态的重建,其作用机制可能通过是减少RhoA基因的蛋白质表达水平而实现的。
Objective To investigate the effect of the sputum drug Baijiezi powder on immune homeostasis in asthmatic rats and RhoA gene expression.Methods Sixty Wistar rats were divided into 6 groups:group A(normal control group),group B(negative control group),group C(asthma model group),group D(dexamethasone group),group E(Baijiezi 2 h group)and group F(Baijiezi 4 h group)(n=10).After successful model being established,rats in group A,group B and group C were normally reared.Rats in group D was intraperitoneally injected with dexamethasone,and the ones in group E and group F were shaved at the back,and were separated by Baijiezi Ai Zhi 2 h and 4 h in both sides of Feishu,spleen and Shenshu.The total number of cells,neutrophils and macrophages,the expression levels of interleukin-6 and tumor necrosis factor(TNF)-α in the bronchoalveolar lavage fluid,and the protein of RhoA gene in lung tissue of rats in the 6 groups were detected.Results HE staining of lung tissue showed that the destruction of alveolar structure and the infiltration of inflammatory cells were significantly alleviated after 2 h and 4 h treatment with dexamethasone and Baijiezi powder.Compared with the normal control group,the number of total cells,neutrophils,macrophages,as well as the TNF-α and IL-6 levels in the bronchoalveolar lavage fluid of rats in the asthma model group were significantly increased(P<0.01).The protein expression level of the RhoA gene in lung tissue also elevated significantly(P<0.01).Treatment with 2 h and 4 h of dexamethasone and Baijiezi powder significantly reduced the number of total cells,neutrophils and macrophages in bronchoalveolar lavage fluid,while the level TNF-α and IL-6 increased significantly(P<0.01).The protein expression level of the RhoA gene in lung tissue significantly increased(P<0.01).Conclusion The sputum drug Baijiezi powder can improve antigen-induced asthma in rats and was helpful to the immune homeostasis.The mechanism may be relatetd to the reduction of the protein expression level of RhoA gene.
