摘要
目的:探究重复经颅磁刺激(Repetitive transcranial magnetic stimulation,rTMS)对缺血性脑卒中急性期小鼠小胶质细胞极化作用的影响。方法:取80只C57BL/6小鼠随机分为假手术组、MCAO对照组、假刺激组及rTMS干预组。rTMS干预组给予高频(10 Hz)刺激,假刺激组给予和rTMS干预组相同线圈环境,但不通电,治疗期间进行转棒实验。疗程结束后制大脑切片行TTC染色计算梗死灶大小,RT-qPCR法检测缺血性脑卒中小鼠小胶质细胞极化相关指标的表达情况。结果:TTC染色结果显示rTMS干预组可减小脑梗死灶体积,并延长转棒实验时间。MCAO对照组iNOS、TNF-α、IL-1β、IL-6表达水平高于假手术组,Arg1、Ym1表达水平低于假手术组,差异有统计学意义(P<0.05);MCAO对照组和假刺激组在各项指标上无明显差异;rTMS干预组的iNOS、TNF-α、IL-1β表达水平低于MACO对照组,而IL-10、Arg1、Ym1表达水平高于MCAO对照组,差异有统计学意义(P<0.05)。结论:rTMS作用于急性期缺血性脑卒中可恢复相关梗死区域的功能,其作用机制可能是通过抑制小胶质细胞M1的极化,促进小胶质细胞M2的极化,降低炎症反应。
Objective:To explore the effect of repetitive transcranial magnetic stimulation(rTMS)on the polarization of microglia in mice with acute ischemic stroke.Methods:Eighty C57 BL/6 mice were randomly divided into sham operation group,MCAO control group,rTMS intervention group and sham stimulation group.The rTMS intervention group was given high-frequency(10 Hz)stimulation.The sham stimulation group was given the same coil environment as the rTMS intervention group,but without electricity.Rotating rod experiments were performed during treatment.After the treatment,the brain slices were stained with TTC to calculate the size of the infarct.And detect the expression of indicators related to the polarization of microglia and neuron in mice with ischemic stroke by Q-PCR method.Results:TTC staining results showed that the rTMS intervention group could reduce the volume of cerebral infarctions.The expression levels of iNOS,TNF-α,IL-1β、IL-6 in the model group were higher than those in the sham operation group,The expression levels of Arg1 and Ym1 in the model group were lower than those in the sham operation group the difference was statistically significant(P<0.05);MCAO control group and sham operation group the stimulation group had no significant difference in various indicators;the expression levels of iNOS,TNF-α,IL-1βin the rTMS intervention group were lower than those in the MACO control group,while the expression levels of IL-10,Arg1,Ym1 were higher than the MACO control group,the difference was statistically significant(P<0.05).Conclusion:rTMS can restore the function of the related infarct area in the acute phase of ischemic stroke.Its mechanism of action may be to inhibit the polarization of microglia M1,promote the polarization of microglia M2,and reduce the inflammatory response.
