卵巢癌组织中酰胺核苷酸反义转氢酶及RNA1表达观察患者预后影响因素分析 被引量:3

摘要 目的观察卵巢癌组织酰胺核苷酸反义转氢酶RNA1(NNT-AS1)的表达变化,分析卵巢癌预后的影响因素。方法82例卵巢癌患者,均行肿瘤切除术,术中保留卵巢癌、癌旁组织,采用实时荧光定量聚合酶链反应检测NNT-AS1,比较不同临床病理特征的卵巢癌患者NNT-AS1表达差异,Kaplan-Meier法绘制不同NNT-AS1表达的卵巢癌患者无进展生存期(PFS)、总生存期(OS)生存曲线。采用单因素和多因素Cox比例风险模型分析卵巢癌患者预后的影响因素因素。结果卵巢癌及癌旁组织NNT-AS1相对表达量分别为0.53±0.19、1.52±0.36,二者比较,P<0.05。低中度分化、Ⅲ~Ⅳ期、CA125≥200 U/mL、ERα阳性患者癌组织NNT-AS1表达低于高度分化、Ⅰ~Ⅱ期、CA125<200 U/mL、ERα阴性者(P均<0.05)。NNT-AS1低表达者PFS生存率、OS生存率分别为27.91%(12/43)、39.53%(17/43),NNT-AS1高表达者分别为56.41%(22/39)、76.92%(30/39),二者比较,P均<0.05。FIGO分期Ⅲ~Ⅳ期、癌组织NNT-AS1相对表达量<0.53与卵巢癌患者的预后不良有关(OR分别为2.065,1.895;95%CI分别为1.989~2.127,1.715~1.953;P均<0.05)。结论卵巢癌组织中NNT-AS1表达降低。卵巢癌预后不良的因素有FIGO分期Ⅲ~Ⅳ期、癌组织NNT-AS1相对表达量<0.53。 Objective To observe the amide nucleotide antisense hydrogenase RNA1(NNT-AS1)expression in ovarian cancer tissues and to analyze the prognostic factors of ovarian cancer.MethodsTotally 82 patients with ovarian cancer were treated with tumorectomy.The ovarian cancer tissues and adjacent tissues were preserved during operation.The real-time fluorescent quantitative polymerase chain reaction(FQ-PCR)was used to detect NNT-AS1.The differences of NNT-AS1 expression in patients with different clinicopathological characteristics were compared.The progression free survival(PFS)and overall survival(OS)curves of ovarian cancer patients with different NNT-AS1 expression were drawn by Kaplan Meier method.The univariate and multivariate cox proportional hazard models were used to analyze the prognostic factors of patients with ovarian cancer.ResultsThe relative expression levels of NNT-AS1 in ovarian cancer tissues and adjacent tissues were 0.53±0.19 and 1.52±0.36,with statistically significant difference between these two groups(P<0.05).The expression of NNT-AS1 in cancer tissues of low and moderate differentiation,stageⅢ-Ⅳ,CA125≥200U/m L and ERαpositive was lower than that of highly differentiation,stageⅠ-Ⅱ,CA125<200 U/m L and ERαnegative(all P<0.05).The rates of PFS and OS were 27.91%(12/43)and 39.53%(17/43)in patients with low expression of NNT-AS1,and the rates of PFS and OS were 56.41%(22/39)and 76.92%(30/39)in patients with high expression of NNT-AS1,with statistically significant difference between these two groups(P<0.05).FIGO stageⅢ-Ⅳand NNT-AS1<0.53 were associated with poor prognosis of ovarian cancer patients(OR=2.065,1.895;95%CI were 1.989-2.127,1.715-1.953,respectively;all P<0.05).ConclusionsThe expression of NNT-AS1 in the ovarian cancer tissues decreases.The factors for poor prognosis of ovarian cancer are FIGO stageⅢ-Ⅳand NNT-AS1<0.53.
