川崎病患儿差异表达基因筛选及相应蛋白表达观察

Screening of DEGs and expression of corresponding proteins in children with KD

目的运用生物信息学方法分析川崎病(KD)患儿差异表达基因(differentially expressed genes,DEGs),并观察KD患儿血清中相应DEGs蛋白的表达变化。方法①KD患儿DEGs筛选:从KD数据库中获取KD的基因组数据,通过R软件中edge R软件包分析KD患儿及健康儿童DEGs。②KD患儿DEGs蛋白表达观察:典型KD患儿40例,健康儿童15例,抽取外周静脉血,采用ELISA双抗体夹心法检测基质金属蛋白酶8(MMP-8)、嗅介蛋白-4(OLFM4)蛋白。结果与健康儿童相比,KD患儿存在288个DEGs,其中表达明显上调基因186个、明显下调基因102个。MMP-8基因和OLFM4基因差异最为明显(P均<0.05)。KD患儿外周血MMP-8、OLFM4蛋白表达水平分别为(73.21±13.66)、(26.37±5.88)ng/mL,健康儿童分别为(46.32±9.38)、(9.37±5.23)ng/mL,二者比较,P均<0.05。结论KD患儿有MMP-8、OLFM4等288个DEGs。KD患儿外周血中MMP-8、OLFM4蛋白表达水平升高。

Objective To analyze the differentially expressed genes(DEGs)in children with Kawasaki disease(KD)based on bioinformatics analysis,and to observe the changes of corresponding PEGs proteins in serum of children with KD.Methods①Screening of DEGs in children with KD:The genomic data of KD were obtained from KD database.The DEGs in children with KD and healthy children were examined by the edge R software package in the R software.②Observation of protein expression in children with KD:40 children with complete Kawasaki disease(CKD)and15 healthy children were randomly selected.The peripheral blood of each participant was collected.ELISA double antibody sandwich method was performed to measure the expression levels of matrix metalloproteinases 8(MMP-8)and olfactomedin-4(OLFM4).ResultsCompared with healthy children,there were 288 DEGs in children with KD,in which 186were up-regulated and 102 were down-regulated.The differences of MMP-8 and OLFM4 were the most significant(both P<0.05).The protein levels of MMP-8 and OLFM4 in peripheral blood of children with KD were(73.21±13.66)ng/mL and(26.37±5.88)ng/mL,while these in healthy children were(46.32±9.38)ng/mL and(9.37±5.23)ng/mL,respectively(both P<0.05).ConclusionsThere are 288 DEGs such as MMP-8 and OLFM4 in serum of children with KD.The expression levels of MMP-8 and OLFM4 in peripheral blood of children with KD increase.
