基于网络药理学探究大黄-苦杏仁药对治疗克罗恩病的作用机制 被引量:3

Network Pharmacology Study on Rhubarb Rhizome-Almonds Amygdalus Couplet in Treatment of Crohn’s Disease

摘要 目的:探讨大黄-苦杏仁药对治疗克罗恩病(crohn's disease,CD)的作用机制。方法:借助TCMSP数据库检索中药大黄、苦杏仁的化学成分和作用靶点,通过UniProt、Malacards和PubMed等数据库查询疾病靶点,进而运用Cytoscape 3.2.1软件构建成分-靶点网络,通过DAVID数据库进行京都基因与基因组百科全书(KEGG)通路富集分析,探究其作用机制。结果:共筛选出46个活性成分,其中OB值(Oral bioavailability)≥30%的活性成分14个(大黄4个,苦杏仁10个),其中12个活性成分参与作用靶点的一级交互网络构建。活性成分的直接作用靶点共4个,分别为白介素-10(IL-10)、肿瘤坏死因子-α(TNF-α)、信号传导与转录激活因子3(STAT-3)、半胱氨酸蛋白酶CPP32(Caspase-3)。KEGG通路富集筛选涉及Th1、Th2、Th17及调节性T细胞(Treg cell),信号通路包括Toll样受体通路、JAK-STAT通路。结论:大黄-苦杏仁药对可通过多成分、多靶点、多通路治疗CD,其作用的物质基础及机制仍需深入发掘。 To probe the mechanism of Rhubarb Rhizome-Almonds Amygdalus Couplet on Crohn’s disease(CD),chemical components and targets of two herb medicines in traditional Chinese medicine system pharmacology platform were selected.UniProt,Malacards,PubMed and DAVID databases as well as Cytoscape 3.2.1 software were applied.46 components were filtered,among them 14 components with Oral bioavailability≥30%(4 Rhubarb Rhizome,10 Almonds Amygdalus),7 components were involved in the construction of the first-level interaction network of the target.There are 4 direct targets for 7 components,including interleukin-10,tumor necrosis factor-α,signaling and transcriptional activator 3,and cysteine protease CPP32.The KEGG pathway enrichment analysis involves Th1,Th2,Th17 and regulatory T cells and the pathway includes Toll-like receptor and JAK-STAT signaling pathways.It is necessary to further study that the material and mechanism of Rhubarb Rhizome-Almonds Amygdalus Couplet treat CD through multiple targets and pathways.
