基于DNA条形码的四川老君山国家级自然保护区尺蛾科昆虫种类鉴定 被引量:1

摘要 为了解四川老君山国家级自然保护区鳞翅目Lepidoptera尺蛾科Geometridae资源,于2019年6—8月和2020年5—6月通过灯诱法采集2402号标本,运用DNA条形码技术对标本进行分子鉴定。通过BOLD数据库,邻接法构建系统发育树和ABGD在线工具对获得的297条线粒体细胞色素C氧化酶亚基Ⅰ(COⅠ)基因序列进行物种界定,结合BOLD和GenBank中70条相关种序列共367条COⅠ基因序列分析遗传距离。发现297条序列隶属于191种,其中95种鉴定到种,35种鉴定到属;鉴定到属或种水平的序列隶属于92属。计算遗传距离发现,种内遗传距离为0~4.36%,平均种内遗传距离为0.63%,种间遗传距离为3.43%~19.12%,平均种间遗传距离为11.91%。结果表明,保护区尺蛾科昆虫种类丰富,基于COⅠ基因的DNA条形码技术能有效区分尺蛾科物种。 To understand the resource of Geometridae(Lepidoptera)in the Laojunshan National Nature Reserve(Sichuan Province),a total of 2402 specimens were collected from June to August,2019 and from May to June,2020.Molecular identification of specimens was carried out by using DNA barcoding technology.Based on the database of BOLD,Neighbor-Joining phylogenetic tree and Automatic Barcode Gap Discovery online tool were used to identify and delimit the 297 mitochondrial cytochrome c oxidase subunitⅠsequences(COⅠ).Additionally,367 COⅠsequences,including 70 sequences obtained from BOLD or GenBank databases,were used for genetic distance analysis.The results showed that the 297 sequences belonged to 191 species of Geometridae,and 95 of them were identified to the species level,and 35 were identified to the genus level.After excluding the sequences identified to the family level,the remaining sequences belonged to 92 genera.Among the 367 sequences,the intraspecific genetic distance was 0-4.36%with an average of 0.63%,and the interspecific genetic distance was 3.43%-19.12%with an average of 11.91%.This study suggested that the species of Geometridae can be well-differentiated by DNA barcoding technology based on COⅠgene,and there were abundant species of Geometridae in the reserve.
