三亚市首例新型冠状病毒肺炎核酸检测实验分析

摘要 目的为确定疑似新型冠状病毒肺炎首例病例是否感染新型冠状病毒。方法采用read-time PCR方法对咽拭子标本检测新型冠状病毒开放读码框(ORF1ab)和核壳蛋白(N)基因,回顾性试剂比对验证所使用的试剂结果是否一致、准确、可靠。结果首例病例为新型冠状病毒开放读码框ORF1ab和核壳蛋白N基因核酸阳性。结论首例病例的核酸检测结果的准确性和及时性,为疫情及时防控提供可靠的科学依据,回顾性试剂比对实验验证了首例病例所使用的新冠病毒核酸检测试剂盒质量是可靠的,结果是准确的。 Objective To confirm whether the first case of suspected the COVID-19 is infected with novel coronavirus.Methods The real-time reverse transcription polymerase chain reaction(RT-PCR)assay was performed to detect Open reading frame(ORF1ab)and nucleocapsid protein(N)genes of novel coronavirus genes in pharyngeal swab specimens,consistency,accuracy and reliability of the results of the reagents used were retrospectively tested,Compared and verifyed.Results The first case was positive for the novel coronavirus ORF1ab and N genes.Conclusion The accuracy and timeliness of the nucleic acid test results of the first case provide a reliable scientific basis for the timely prevention and control of the epidemic.The retrospective reagent comparison experiment verify that the quality of the Diagnostic Kit for the COVID-19 ORF1ab/N(Fluorescence PCR)used in the first case is reliable and the results are accurate.
