沙棘提取物延缓糖尿病大鼠白内障进展作用的研究 被引量:1

摘要 目的:探讨沙棘提取物(sea buckthorn,SBT)对糖尿病大鼠白内障的作用。方法:随机将40只雄性SD大鼠分成正常组、糖尿病(diabetic mellitus,DM)组、低SBT组、高SBT组,每组10只。腹腔注射1%链脲佐菌素造模,每4周检测大鼠血糖,拍摄大鼠晶状体眼前节图像,12周时取右眼晶状体行HE染色,采用比色法取左眼晶状体检测一氧化氮(nitric oxide,NO)、丙二醛(malondialdehyde,MDA)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-PX)的水平;采用酶标仪检测大鼠血清中甘油三酯(triglyceride,TG)、总胆固醇(total cholesterol,TC)、低密度脂蛋白(low density lipoprotein,LDL)的水平。结果:分别于1周、4周、8周、12周时监测大鼠血糖,同一组大鼠不同时间的血糖比较,差异无统计学意义(P>0.05),DM组血糖与低SBT组和高SBT组比较,差异有统计学意义(P<0.05)。12周时正常组无晶状体混浊,其余3组全部出现>Ⅲ级的晶状体混浊,而低SBT组和高SBT组晶状体的混浊程度与DM组相比明显降低,差异有统计学意义(P<0.05,χ^(2)=52.57)。12周时低SBT组和高SBT组NO、MDA、GSH-PX的水平与DM组相比均明显降低,差异有统计学意义(P<0.05)。低SBT组和高SBT组与DM组晶状体中的LDL相比均明显升高,差异有统计学意义(P<0.05);12周时低SBT组和高SBT组晶状体中的TG、TC水平与DM组相比,差异无统计学意义(P>0.05)。结论:SBT能延缓DM大鼠白内障的进展,这可能与SBT降血脂、降血糖及发挥抗氧化应激的作用有关。 Objective:To study the effect of sea buckthorn(SBT)extract on retarding cataract progression in diabetes mellitus(DM)rats.Methods:Forty male SD rats were randomly divided into normal group,DM group,low SBT group and high SBT group,10 rats in each group.The blood glucose and the image of anterior segment of lens were measured every 4 weeks,and the right eye was stained with HE,and the left eye was taken to detect the levels of nitric oxide(NO),malondialdehyde(MDA)and glutathione peroxidase(GSH-PX)in the lens at 12 weeks.At the same time,the serum levels of triglyceride(TG),total cholesterol(TC)and low density lipoprotein(LDL)were measured.Results:The blood glucose of rats was monitored at 1 week,4 weeks,8 weeks and 12 weeks.There was no significant difference in blood glucose at different time in the same group(P>0.05).Blood glucose in DM group was significantly different from that of low SBT group and high SBT group(P<0.05).At the 12th week,there was no lens opacity in the normal group,lens opacity of gradeⅢor above was found in other groups,and the lens opacity in the low SBT group and high SBT group was significantly less than those in the DM group(P<0.05,χ^(2)=52.57).At the 12th week,the level of NO,MDA and GSH-PX in low SBT group and high SBT group were significantly lower than those in DM group.The level of LDL in low SBT group and high SBT group were significantly higher than those in DM group,but there was no significant difference in serum TG and TC between low SBT group,high SBT group and DM group at 12 weeks.Conclusion:SBT extract can delay the progression of cataract in DM rats by reducing the levels of LDL,blood glucose,NO,MDA and playing an anti-oxidative role.
