摘要
目的探讨Polo样激酶1(PLK1)在哮喘发病机制中的作用。方法采用大鼠哮喘模型,随机分成哮喘组、对照组和地塞米松组,免疫组织化学法和实时定量PCR法检测肺组织PLK1的表达。结果阳性细胞主要表达在支气管上皮细胞和肺泡上皮细胞的胞浆中。哮喘组肺组织PLK1蛋白和mRNA的表达水平显著高于对照组(P<0.01);PLK1蛋白和mRNA的表达水平在地塞米松组与哮喘组中差异均无统计学意义(P>0.05)。肺组织PLK1蛋白和PLK1 mRNA的表达水平呈显著正相关(r=0.38,P<0.05)。结论哮喘大鼠肺组织PLK1的表达增强,表达升高了的PLK1可能在哮喘发病机制中起到了促进炎症的作用,而地塞米松对PLK1的表达却没有影响。
Objective To investigate the potential roles of Polo like kinase 1(PLK1) at rat asthma pathogenesis.Methods Rat asthma models were divided into asthma group,control group and desamethasome group,and the levels of PLK1 protein and m RNA were detected by immunohistochemical method and Real-time PCR method.Results The positive cells were mainly expressed in the cytoplasm of bronchial epithelial cells and alveolar epithelial cells.The levels of PLK1 protein and mRNA in lung tissue of asthma group were significantly higher than those of control group(P<0.01).There was no statistical significance on the difference in the levels of PLK1 protein and m RNA between dexamethasone group and asthma group(P>0.05).The levels of PLK1 protein and PLK1 mRNA in lung tissues were significantly positively correlated(r=0.38,P<0.05).Conclusion The expression of PLK1 in the lung tissue of rats with asthma was enhanced,and the elevated expression of PLK1 may play a role in promoting inflammation in the pathogenesis of asthma,while dexamethasone had no effect on the expression of PLK1.
